Establishment of HPMEC Inflammation Model

Human pulmonary microvascular endothelial cells (HPMECs) were purchased from Sunncell Biotech Co., Ltd. (China). These cells were cultured in DMEM medium (Dalian Meilun Biotech Co., Ltd., China) supplemented with 10% fetal bovine serum (FBS) and 100 U/mL penicillin and 100 μg/mL streptomycin and grown at 37 °C in a 5% CO2 incubator. Cells were cultured to 80% confluency and used for experiments in this study. Trypsin (Dalian Meilun Biotech Co., Ltd., China) was used to passage cells.
According to previous studies [20 (link),21 (link)], cells were treated with LPS (1 mg/L) or the same dose of saline for 24 h and then collected. Huzhou Hippo Biotechnology Co., Ltd. (China) created inhibitors of miR-346 (miR-346 inhibitor) and negative controls (nc inhibitor), small interfering RNA targeting BCL6 (si-BCL6) and negative controls (si-NC).These inhibitors were transfected into HPMECs using Entranster™-R (Entrangreen Biosystem, Ltd., China) in strict accordance with the manufacturer's guidelines.
