Evaluating UCA1 and miR-331-3p in U2932 cells

U2932 cells (4×105) in the logarithmic growth phase were seeded into a six-well plate. When cell confluence reached 70–80%, U2932 cells were transfected with pcDNA3.1-UCA1(UCA1 vector, 40 nM), small interfering (si)-UCA1 (40 nM), miR-331-3p mimics (40 nM), miR-331-3p inhibitor (80 nM), or their corresponding negative control (40 nM) by Lipofectamine 2000 (Invitrogen; Thermo Fisher Scientific, Inc.). Transfection of miRNA Mimics and Inhibitors

Negative control (NC) mimics, miR-331-3p mimics, and miR-inhibitors were designed and synthesized by Genepharm (Shanghai, China). Cells were transfected with NC mimics, miR-331-3p mimics or miR-inhibitors using transfection reagent (pufei biotech, China) following the manufacturer’s instructions.

Validating miR-331-3p and MLLT10 Interaction

The online software miRDB (http://mirdb.org/index.html) was used to predict the target gene of miR-331-3p. Dual-luciferase reporter assay was used to identify the correlation between miR-331-3p and its target gene MLLT10. Briefly, 293A cells, MLLT10-wt plasmids, and MLLT10-Mut plasmids were purchased from HonorGene. MiR-331-3p mimics and mimic NC were purchased from Shanghai GenePharma Co., Ltd. The MLLT10-wt or MLLT10-Mut or MiR-331-3p mimics or mimic NC were cotransfected into precultured 293A cells using Lipofectamine 2000. After 48 h, the luciferase activity was analyzed with the Dual Luciferase Reporter Assay System (Promega, USA).
