Immunofluorescence Staining of Exosome Markers

Cells were grown on glass dishes for 24 h then fixed for 15 min in 4% paraformaldehyde and permeabilized with 0.3% Triton X-100 for 15 min at room temperature. Next, cells were blocked with 10% goat serum for 1 h at room temperature, followed by overnight incubation with primary antibodies at 4°C. After being washed with PBS, the cells were incubated with Alexa-conjugated secondary antibody (Cell Signaling Technology, Inc.) at 37°C for 1 h and then incubated with DAPI at 37°C for 5 min. The cells were visualized and images captured on a confocal microscope (Olympus LV3000; Olympus Corporation). Primary antibodies were: RPL9 (dilution: 1:50; Abcam; cat. no. ab182556), TSG101 (dilution: 3:100; MilliporeSigma; cat. no. SAB2702167), VPS4A (dilution: 1:50; Santa Cruz Biotechnology; cat. no.
