Cytokine Profiling of HIV-specific CD8+ T cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
One million CD8 T cell-depleted PBMCs (Dynabeads CD8, StemCell) were incubated with an HIV Gag peptide pool (1μg/ml/peptide) or left unstimulated and incubated at 37°C in RPMI supplemented with 10% HAB and PenStrep. Cytokine secretion in supernatants was measured 48 hours after stimulation on a Luminex beads array platform, as previously described19 (link), 20 (link). Measurements were performed in duplicates using the Human High Sensitivity Cytokine Premixed Kit B (R&D Systems) for IL-2, IFN-γ, TNF-α, IL-17A, IL-22 and IL-31 or for IL-17F on a Bio-plex 200 array system (Bio-Rad Laboratories) per manufacturer’ s instructions. CXCL13 was measured separately with the Human CXCL13 Quantikine ELISA Kit (R&D Systems), as per manufacturer’s instructions.

Multiplex Plasma Protein Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
To expand the range of proteins detected with the previous method, the concentration of soluble inflammatory markers in plasma was analysed using three multiplex bead-based immunoassays. These cover a wider range of cytokines, chemokines, growth factors and acute-phase protein biomarkers. Globally, 63 distinct plasma proteins (76 analytes due to some duplications across kits) were assessed simultaneously, using the Bio-Plex 200 array system according to the manufacturer's instructions (Bio-Rad). Further details on immune-based multiplex assays and methodology for immunoglobulin quantification, as well as a reduced validation of our proteomic results using ELISA can all be found in the supplementary material.

Multiplex Cytokine Profiling in Plasma

Check if the same lab product or an alternative is used in the 5 most similar protocols
The concentrations of circulating cytokines, chemokines, and other analytes were assayed using bead-based multiplex technology. Irradiated plasma samples were incubated with magnetic beads from Milliplex NHP cytokine premixed 23-plex panel (EMD Millipore, Billerica, MA) kits according to the recommendations provided. Analytes measured included IL-1β, IL-1 receptor antagonist (IL-1RA), IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, IL-12/23 (p40), IL-13, IL-15, IL-17, IL-18, gamma interferon (IFN-γ), granulocyte colony-stimulating factor (G-CSF), granulocyte–macrophage colony-stimulating factor (GM-CSF), monocyte chemoattractant protein 1 (MCP-1), macrophage inflammatory protein 1α (MIP-1α), MIP-1β, tumor necrosis factor alpha (TNF-α), transforming growth factor alpha (TGF-α), soluble CD40 ligand (sCD40L), and vascular endothelial growth factor (VEGF). The concentrations in each plasma sample were measured using a Bioplex-200 array system (Bio-Rad, Hercules, CA).

Multiplex Profiling of Plasma Cytokines and Coagulation Markers

Check if the same lab product or an alternative is used in the 5 most similar protocols
Concentrations of cytokines and other analytes were assayed using bead-based multiplex technology. Irradiated plasmas were incubated with magnetic beads from Milliplex NHP Cytokine Premixed 23-plex Panel (EMD Millipore, Billerica, MA) or ProcartaPlex NHP TGF-beta 1 simplex (eBioscience, Vienna, Austria) kits according to the manufacturer protocols. The concentrations in each plasma sample were measured using a Bioplex-200 array system (BioRad, Hercules, CA).
Plasma levels of coagulation-associated parameters were quantified using a flow-based Legendplex Human Thrombosis 7-plex bead immunoassay (Biolegend, San Diego, CA). Samples were processed in duplicate following the kit instructions to measure concentrations of the following analytes: p-selectin, d-dimers, p-selectin glycoprotein ligand-1 (PSGL-1), tissue plasminogen activator (tPA), soluble CD40-ligand (sCD40L), plasminogen activator inhibitor-1 (PAI-1), and factor IX. Approximately 2,100 events (beads) per sample were collected on a FACS Canto II cytometer (BD Biosciences, San Jose, CA) using BD FACS Diva software and the raw data was analyzed with BioLegend’s cloud-based LEGENDplex™ Data Analysis Software.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
