Intracellular Cytokine Staining in T Cells

For detection of intracellular cytokines in T cells, lymphocytes isolated from the spleen and lung were stimulated in vitro with peptides including GP33-41 and GP276-286 (0.2µg/ml) in the presence of golgi-stop, golgi-plug, and anti-CD107a (1D4B) (BD Biosciences) for 5 h. Stimulated lymphocytes were permeabilized with Cytofix/Cytoperm (BD biosciences) and then stained with the following monoclonal antibodies (BD Biosciences): anti-IFN-γ (XMG1.2), anti-TNF-α (MP6-XT22), and anti-IL-2 (JES6-5H4).
