Serum Lipid and Bile Acid Profiling

The levels of total triglyceride (TG), total cholesterol (TC), high-density-lipoprotein cholesterol (HDLC), low-density-lipoprotein cholesterol (LDLC), and total bile acid (TBA) in serum were measured by analysis kits (A110-2, A111-2, A112-1, A113-1, and E003-2-1, Jiancheng Institute of Biotechnology, Nanjing, China). Non-esterified fatty acid (NEFA) was measured by an automatic biochemical analyzer (Olympus Au5400). The concentrations of very-low-density-lipoprotein-cholesterol (VLDL-C) and lysophosphatidylcholine (LPC) were measured by enzyme-linked immunosorbent assay kits (H249, Jiancheng, Nanjing and CEK621Ge, Cloud-clone corp, Wuhan, China, respectively) according to the instruction manual.

Serum and Hepatic Lipid Profiling

Serum parameters including total cholesterol, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol levels were measured with the commercial kits (A111-2, A112-2, and A113-2; Jiancheng). Serum chemistry tests including alanine aminotransferase, aspartate aminotransferase, creatine kinase, lactate dehydrogenase, urea nitrogen, creatinine, uric acid, albumin, and total protein were performed with the commercial kits (MAK052, MAK055, MAK116, MAK066, MAK006, MAK080, MAK077; 09753 and 71285-M; Sigma–Aldrich). Triglyceride was extracted using 5% NP-40 solution in liver and heated at 90 °C for twice. Then it was cooled down to room temperature and centrifuged at 12,000 rpm for 2 min to collect transparent supernatant. The serum and hepatic levels of triglyceride were measured using TG kit (K952; BioVision).

Metabolic Assessment in Fasted Mice

Mice were fasted for 16 h before the tests. For the glucose tolerance tests (GTT), 2 g/kg glucose was injected intraperitoneally and plasma glucose levels were measured before the injection and at 15, 30, 60, 90, 120 and 150 min after the injection. For insulin tolerance tests (ITT), 0.75 U/kg insulin was given intraperitoneally and glucose levels in plasma were also determined before the injection and at 15, 30, 60, 90, 120 and 150 min after the injection. Fasting insulin levels was detected by an ELISA kit (EZRMI-13K, Millipore, Billerica, MA). Homeostasis model assessment of insulin resistance (HOMA-IR) was calculated as fasting insulin concentration (mU/L) × fasting glucose concentration (mmol/L)/ 22.5. Hepatic and plasma triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and alanine aminotransferase (ALT) were measured by enzymatic assay kits (A110-2, A111-2, A113-2, C009-2 Nanjing Jiancheng Bioengineering Institute, China). Serum levels of calcium and phosphate was determined by spectrophotometry (BC1655, BC0720, Beijing Solarbio Science & Technology Co. Ltd, China).
