Formulation of Monocaprin-based Gel

A monocaprin stock of 100 g/L in propan-1,2-diol (BDH Chemicals, VWR International) was made. The required amount of carboxymethylcellulose (CMC) sodium salt (medium viscosity, 400–800 cP, 2% in H2O at 25 °C, Sigma-Aldrich) was added to half the required final volume of sterile deionised water (i.e. for 100 g formulation, 50 ml of water is added) in a sterile conical flask and placed on an orbital shaker at 200 rpm overnight at room temperature. A double concentrate of polysorbate 20 (BDH Chemicals, VWR International) was made up in sterile deionised water. The monocaprin stock was then added at double the required overall concentration and extra propan-1,2-diol was added (if required) to bring the concentration up to 10%. After briefly mixing this solution, the double concentrate of the polymer was added to the double concentrate of polysorbate 20, monocaprin, and propan-1,2-diol to the required weight. This thickened solution was mixed at room temperature on a Labnet Mini LabRoller (Edison, NJ, United States) with a 5 × 50 ml tube rotisserie.

Peanut Cultivars Analysis by MALDI-MSI

Acetonitrile and methanol were purchased from Merck (Darmstadt, Germany). MilliQ water (Millipore, Bradford, PA, USA) was used in all experiments. Carboxymethyl cellulose (CMC) sodium salt, formic acid (FA), and 2,5-Dihydroxybenzoic acid (DHB) were purchased from Sigma-Aldrich (St. Louis, MO, USA). All reagents and solvents used in this study were of analytical grade.
Three peanut cultivars with different testa colors were used in this study. “Zhonghua 9” (ZH9) is a black peanut, “Kangqibaihong” (KQBH) has white testa, and “Huapi” (HP) has a red seed coat with white spot (Figure 1a). The peanut seed structure was mainly distinguished as embryo, cotyledon, and testa. From a histological point of view, the embryo was divided into three parts, including radical, plumular axis, and plantule (Figure 1b). All plant materials were grown in the test field of Oil Crop Research Institute, Chinese Academy of Agricultural Sciences, Wuhan, China. The peanuts were harvested at the mature stage (about 120 days after planting), and seeds with similar size (about 2 cm in longitudinal direction) were collected (Figure 1c). After removing testa, seeds were immersed with 10% gelatin (wt/vol) solutions in an embedding box, then frozen on dry ice and stored in a −80 °C refrigerator until use for MALDI-MSI analysis.

Hepatic Stellate Cell Isolation and Liver Injury Induction in Rats

Rats were anaesthetized by intraperitoneal injection of 0.1% sodium pentobarbital (40 mg/kg). Subsequently, the rats were subcutaneously injected with 40% CCl4 (Beijing Beihua Fine Chemicals Co., China) dissolved in olive oil suspension (Beyotime Biotechnology, Jiangsu Province, China; 2 ml/kg twice a week) and intragastrically administer with 0.5% carboxymethylcellulose (CMC) sodium salt (Sigma-Aldrich, St. Louis, MO) once a day for 12 weeks with or without RSV (Sigma-Aldrich) daily treatment intragastrically for 12 weeks. The control group was treated olive oil and was orally administered sodium CMC. In some experiments, the ERS inhibitor, 4-PBA (Sigma-Aldrich), was intraperitoneally administrated (10 mg/kg). At the end of the 12 weeks, rats were anesthetized with 40 mg/kg sodium pentobarbital, and 5–10 ml blood was collected from the heart; following blood collection, rats were euthanized by cervical dislocation, and liver tissue samples were collected. To isolate HSCs in the indicated time, livers were digested by collagenase/pronase. The HSCs were obtained from the non-parenchymal cell fraction by density-gradient centrifugation using 28% Nycodenz (Nycomed Pharma, Oslo, Norway) and fluorescence-activated cell sorting using their typical retinoid fluorescence.
