Murine Obesity Models for ex vivo Analysis

C57BL/6J mice and OT-II (B6.Cg-Tg(TcraTcrb)425Cbn/J) transgenic mice were purchased from the Jackson Laboratory (New Jersey, USA) and housed at the animal facilities of Weill Cornell Medicine (WCM) according to IACUC regulations. All animal protocols were approved by the WCM IACUC. Studies were conducted using female and male mice. For female mice, C57BL/6J mice (at 10 weeks of age) were fed with a low-fat diet (LFD, 10% fat, Research Diets, D12450Bi) or a high-fat diet (HFD, 60% fat, Research Diets, D12492i) for 18–25 weeks before sacrifice for subsequent ex vivo experiments. Regarding male mice, DIO control (JAX #380056) or DIO (JAX #380050) mice in the C57BL/6J background were purchased from Jackson Laboratory (New Jersey, USA) and fed either a low-fat diet (LFD, 10% fat, Research Diets, D12450Bi) or a high-fat diet (HFD, 60% fat, Research Diets, D12492i), respectively, up to 25 weeks before sacrifice for subsequent ex vivo experiments.

High-Fat Diet Feeding in Mice

Dietary Obesity Induction in Mice

All mouse care and experimental procedures were approved by the institutional animal care research advisory committee of the Albert Einstein College of Medicine. All experiments were performed in accordance with relevant guidelines and regulations. Mice used in experiments included C57BL/6J mice (The Jackson Laboratory, stock # 000664) and C57BL/6J DIO mice used as noted (the Jackson Laboratory, Stock # 380050). Both male and female mice were used for most experiments and were maintained with a 12 hours light-dark cycle. C57BL/6J mice at 5 weeks of age were fed a low fat diet (LFD, 70% calories provided by carbohydrates, 20% by protein, and 10% by fat; 3.85 kcal/g, D12492J, Research Diets), or a high fat diet (HFD, 20% calories by carbohydrate, 20% by protein, and 60% by fat; 5.21 kcal/g, D12492; Research Diets) with ad libitum access to water for 5 weeks at 28°C. Mice were euthanized by an overdose of Isoflurane.

High-Fat Diet Mouse Model for Obesity

Male C57BL/6J wild-type (WT) mice were obtained from The Jackson Laboratory (Bar Harbor, ME). Mice were fed normal chow diet (ND; 12% kcal from fat; Picolab Rodent Diet 5010, Purina Mills) after weaning until 8 weeks old. Starting at 8 weeks of age mice were fed western high-fat diet (HFD) (41% kcal from fat; Dyet 112734, Dyets Inc.) for 2–24 weeks or maintained on ND for the same period. CD11a-deficient (CD11a−/−)42 (link) and C57BL/6NHsd control mice (Harlan Laboratories) were fed ND or HFD for 16 weeks. Animal studies and procedures were approved by the Institutional Animal Care and Use Committee of Baylor College of Medicine. For micro–computed tomography (micro-CT) studies, C57BL/6J mice fed ND or 60%HFD (60% kcal fat; D12492, Research Diets) for 12 weeks were donated by Willa Hsueh (Houston Methodist Research Institute, Houston, TX). No randomization was used to allocate the animals to experimental groups. The investigator was not blinded to the group allocation.

Phloroglucinol Effects on HFD Mice

C57BL/6J and C57BL/6N male mice were obtained from OrientBio (Kyounggido, Korea). Mice were housed at the animal facility of the Lee Gil Ya Cancer and Diabetes Institute (CACU, Gachon University, Incheon, Korea), in accordance with “Guidelines and Animal for Users”. Six week-old male C57BL/6J mice were fed an HFD (60% fat primarily from lard, Research Diets, Inc., New Brunswick, NJ, USA, #D12492). After 10 weeks (at 16 weeks of age), mice were randomly divided into two groups: the PBS-treated HFD group (HFD + PBS; n = 5) and the phloroglucinol-treated HFD group (HFD + phloroglucinol; n = 5). Phloroglucinol (100 mg/kg in PBS) or PBS was administered by oral intubation daily for 9 weeks. Body weight and food consumption were measured weekly.

Genetic Mouse Models of Liver Fibrosis

Male 8- to 10-week-old C57BL/6J mice were obtained from The Jackson Laboratory. Mice were maintained under a 12-hour-light/12-hour-dark cycle and fed ad libitum. To induce liver fibrosis, 10- to 12-week-old mice were subjected either to BDL surgery or CCl4 treatment as indicated. To generate diet-induced obesity, C57BL/6J mice were fed a high-fat diet (D12492, Research Diets Inc., 60% of calories as fat). Lean controls were mice kept on a standard laboratory diet (NIH-31 rodent diet) for 14 weeks. Cnr1−/− mice were generated as described previously (66 (link)). Nos2−/− mice were purchased from The Jackson Laboratory. Cnr1−/−, nos2−/−, and mdr1a−/− mice were on C57Bl/6J genetic background.

Obesity Induction in Transgenic Mice

Female h-NAG-1 transgenic mice and wild type C57BL/6J mice at 5 weeks of age from a colony known to be positive for Helicobacter were placed on either 10% fat diet (D12450B) or 60% fat diet (D12492, Research Diets, New Brunswick, NJ) for 20 weeks. Isolation and Characterization of Adipocyte-Derived Extracellular Vesicles

Male C57BL/6 J mice were purchased from Janvier Labs at an age of 6-8 weeks. Upon arrival, mice were group-housed and kept in a constant environment with the ambient temperature set to 22 ± 2 °C with constant humidity (45–65%) and a 12 hrs/12 hrs light/dark cycle. Murine AdEVs were isolated from the adipocyte fraction of the epididymal white adipose tissue (eWAT) of male C57BL/6 J mice fed with high fat diet (HFD, #D12332, Research Diets Inc) or chow (Altromin #1310) for 4–6 months. Aging and Obesity in Mouse Models

Six to 8 weeks old female heterozygous Hsd: Athymic Nude-Foxn1nu/Foxn1+ mice were purchased from Envigo (Indianapolis, IN). Six to 8-weeks-old male C57BL/6J mice were purchased from Jackson Laboratories (Bar Harbor, ME). Mice were housed at the Mayo Clinic animal care facility, which is accredited by the Association for Assessment and Accreditation of Laboratory and Animal Care International (AAALACI). Aging was induced in two separate cohorts of C57BL/6J mice [fed with regular diet or high-fat diet (D12492, Research diets)] by keeping them in-housed for 24 months (24 mo) at the Mayo Clinic animal care facility, i.e., a small cohort of 5 mice, 2 months old was maintained for 22 months fed throughout on regular diet to obtain an aged mice group (24 mo), and, another cohort of 5 mice (2 months old) was fed on high-fat-diet (HFD) to induce obesity and continued on HFD for 22 months to obtain an aged-obese mice group (24 mo). All animal procedures were performed with proper animal handling, adhering to the National Institutes of Health (NIH) guidelines and protocols approved by the Institutional Animal Care and Use Committee (IACUC) at Mayo Clinic, Rochester.
