Trem2 Conditional Knockout in AD Mice

Trem2-/- (Trem2 KO) mice were purchased from Jackson Laboratory, USA (C57BL/6J-Trem2em2Adiuj/J). Trem2flox/flox mice (C57BL/6J-Trem2em2(flox)Smoc, Shanghai Model Organisms Center Inc., China) were mated with Ctsk-Cre knock-in mice (C57BL/6J-Ctskem1(2A-Cre-Wpre-pA)Smoc, Shanghai Model Organisms Center) in which the Cre recombinase gene was specifically expressed in osteoclasts. Ctsk-Cre+/−, Trem2flox/flox (Trem2 cKO) and Ctsk-Cre−/−, Trem2flox/flox (normal littermates) mice were generated by mating Ctsk-Cre−/−, Trem2flox/flox male mice with Ctsk-Cre+/−, Trem2flox/+ female mice. Heterozygous Tg2576 mice (APP+/-) overexpressing the human Swedish APP mutation (B6; SJL-Tg(APPSWE)2576Kha, Taconic Biosciences, Germany) on a mixed C57BL/6×SJL background were crossed with Ctsk-Cre+/-, Trem2flox/flox mice to generate three groups of mice: APP-/-, Ctsk-Cre-/-, Trem2flox/flox (normal littermates); APP+/-, Ctsk-Cre-/-, Trem2flox/flox (Tg2576); APP+/-, Ctsk-Cre+/-, Trem2flox/flox (Tg2576×Trem2 cKO). In the current study, 2-month-old male mice were used for animal experiments with at least three biological repeats in each group. The mice were maintained in accordance with the National Institutes of Health's Guide for the Care and Use of Laboratory Animals, and the study protocols were approved by The Animal Care and Use Committee of Tongji University.

ACL Injury Model in Mice

10-week-old male C57Bl/6J (BL6) (Jackson Laboratory Bar Harbor, ME, USA; Stock No: 000664) or Trem2-/- mice (C57BL/6J-Trem2em2Adiuj/J; Jackson Laboratory Bar Harbor, ME, USA; Stock No: 027197) were subjected to an ACL injury using a non-invasive single dynamic tibial compressive overload model previously described by Christiansen et al. (21 (link)). Briefly, the mouse knee was placed between two vertically aligned plates positioned using an electromagnetic material testing system (ElectroForce 3200, TA Instruments, New Castle, DE, USA) and a compressive force was applied (21 (link)). ACL rupture occurred after a total compressive force, between 12N-18N, was administered. For pain relief, buprenorphine (0.01 mg/kg) and sterile saline were administered intraperitoneally immediately post-injury. All animal experiments were approved by the Lawrence Livermore National Laboratory and University of California, Davis Institutional Animal Care and Use Committee and conformed to the Guide for the care and use of laboratory animals.

Transgenic Mouse Strains for Alzheimer's Research

Wild-type C57BL/6J, B6;FVB-Tg(Aldh1l1-EGFP/Rpl10a)JD130Htz/J, C57BL/6J-Trem2em2Adiuj/J, and C57BL/6-Tg(UBC-GFP)30Scha/J mice were purchased from The Jackson Laboratory. 5xFAD mice1 (link) were purchased from the Jackson Laboratory (MMRRC) and backcrossed onto the C57BL/6J background more than 10 generations before being crossed with other strains. Il3−/− mice on the C57BL/6J background were bred in-house2 (link),3 and crossed with 5xFAD mice. For RNAseq studies, Trem2−/− mice4 (link) on the C57BL/6J background were generated at Washington University School of Medicine, bred in-house, and crossed with 5xFAD mice. Age- and sex-matched animals were used. If sex of the animals is not specifically indicated, groups were sex balanced. Where appropriate, animals were randomly assigned to interventions. All mice were group housed under standard conditions with free access to food and water. All animal protocols were approved by the Animal Review Committee at the Massachusetts General Hospital (protocol no. 2011N000035 and 2015N000044) and were in compliance with relevant ethical regulations.
