DAPI Staining for Apoptosis Analysis

DAPI stain (Roche Applied Science, Rotkreuz, Switzerland) was used to evaluate drug-associated apoptosis, according to the manufacturer's protocol. Transfected and non-transfected H1299 cells (80% confluent) were seeded onto the slides in 6-well plates overnight and exposed to 20 µmol/l CDDP for 48 h at 37°C. Next, the cells were fixed with 4% paraformaldehyde at room temperature for 30 min and stained with 0.5% DAPI for 15–20 min. The stained cells were visualized at ×100 magnification and images were captured using an inverted fluorescence microscope (Olympus BX-51; Olympus Corporation).
