Viral Infection and dsRNA Detection

HeLa cells were mock- or virus-infected with either wt VACV, ΔE3, E3Δ37N, E3Δ37NIBTR−3, E3Δ37NIBTR-7, VACV-A24R-R1, or MPXV. At 6 hpi, cells were trypsinized and fixed with CytoPerm/Fix kit (BD). Cells were stained with rabbit anti-VACV (Abcam) and mouse anti-dsRNA (J2 clone, Scicons) overnight at 4°C. Cells were washed and stained with anti-rabbit-IgG Pacific Blue (BD) and anti-mouse-IgG 488 (Invirtogen) for 4 °C. Cells were washed, resuspended in FACS buffer (2% BSA, 0.9% sodium azide in PBS) and were analyzed by flow cytometry using a BD FACSymphony™ A3 Cell Analyzer and cells were gated to exclude doublet and non-viable cells.
