Conditional Astrocytic Aromatase Knockout

We created the Cyp19a1 Cre/LoxP conditional KO mice with an inducible GFAP promoter to conditionally inactivate the aromatase in their astrocytes. GFAP-Cre/ERT2 (Jackson lab, Bar Harbor, ME, USA. stock no. 012849) mice were crossed with Cyp19a1 flox/flox animals, which had LoxP sites surrounding exons 10 and 11 of the Cyp19a1 gene. Using the methods previously reported, Cyp19a1 flox/flox (FLOX) mice were produced [34 (link)]. We confirmed the genotyping of the animals by a genomic DNA PCR, using the following primers: FLOX-forward: 5′-GCCATATTCCTGCAACAGTTTATTTGAGG-3′; FLOX-reverse: 5′-GTAAAACCATTCCTGGAAAATTCATAACAACC-3′; CreERT2-forward: 5′-GGCCAGGCTGTTCTTCTTAG-3′; CreERT2-reverse: 5′-ATACCGGAGATCATGCAAGC-3′; Internal positive control forward: 5′-CTAGGCCACAGAATTGAAAGATCT-3′; and Internal positive control reverse: 5′-GTAGGTGGAAATTCTAGCATCATCC-3′. The mice with the genotype of Cyp19a1flox/flox; GFAPCreERT2 were identified as the GFAP-ARO-iKO mice. All the animal experiments were approved and authorized by the Institutional Animal Care and Use Committees of Augusta University and UT Health San Antonio.

Conditional Knockout Mouse Models for Cell Lineage Tracing

NestinCreERT2 (Jackson ID #016261), floxed Ednrb (Jackson ID #011080), PdgfraCreERT2 (Jackson ID #018280), Rosa26YFP (Jackson ID #006148), GFAPCreERT2 (Jackson ID #012849), and C57bl/6n (Jackson ID #000664) mice were purchased from the Jackson Laboratory. Floxed ET-1 mice were obtained from Dr. Ralph Shohet at the University of Hawaii as previously described15 (link). All mouse colonies were maintained in the animal facility of Children’s National Hospital, under standard conditions (65–75 °C, 40–60% humidity, 12 h light/dark cycles). All animal procedures were performed ethically, and according to the Institutional Animal Care and Use Committee of the Children’s National Hospital (protocol #30578) and the Guide for the Care and Use of Laboratory Animals (National Institutes of Health).
