Dopamine Receptor Ligand Preparation

Dopamine hydrochloride, (S)-(−)-sulpiride, dimethyl sulfoxide (DMSO), and droperidol were all acquired from Sigma-Aldrich. Haloperidol and piribedil were both acquired from TCI Chemicals. Bromocriptine mesylate was acquired from VWR. Apomorphine hydrochloride was kindly provided by EVERPharma AT GmbH within the context of a different project. Tested compounds (shown in Table S9) were acquired either from SPECS (https://www.specs.net/, accessed in April 2021) or Maybridge (https://www.thermofisher.com/at/en/home/industrial/pharma-biopharma/drug-discovery-development/screening-compounds-libraries-hit-identification.html, accessed in April 2021). All the compounds were dissolved in DMSO, aliquoted, and stored at −80 °C until further use.

Binding Assay for Antipsychotic Drugs

Olanzapine, nemonapride, remoxipride hydrochloride, flupenthixol dihydrochloride, paliperidone, amisulpride, melperone hydrochloride, clozapine, raclopride, domperidone, asenapine maleate and haloperidol hydrochloride used for competition assays were obtained from Tocris Bioscience (Avonmouth, Bristol).

Cardiac Toxicity Evaluation of hERG Inhibitors

Drug assays were performed when the rhythmic beating profiles of the cardiomyocytes became stable. For this study, three typical hERG K+ channel inhibitors, namely, astemizole (Sigma-Aldrich, USA), droperidol (Sigma-Aldrich, USA), and sertindole, were applied at various doses (16 nM, 80 nM, 400 nM, 2 μM, and 10 μM) to treat the cardiomyocytes. These drugs were prepared in dimethylsulfoxide (DMSO, Sigma-Aldrich, USA) and PBS and then prewarmed in a 37 °C water bath before being added to the culture wells. Mechanical beating signals were recorded before drug treatment as controls.

Antipsychotic Drug Cytotoxicity Evaluation

The main chemicals used in this study, including FGAs (chlorpromazine, chlorprothixene, clothiapine, droperidol, flupentixol, fluphenazine, haloperidol, levomepromazine, loxapine, pimozide, prochlorperazine, sulpiride, thioridazine, and trifluoperazine) and SGAs (amisulpride, aripiprazole, brexpiprazole, clozapine, lurasidone, olanzapine, paliperidone, quetiapine, risperidone, ziprasidone, and zotepine) were acquired from Sigma-Aldrich (St. Louis, Missouri, USA) and were of highest purity grade. Before usage, the compounds were dissolved in dimethyl sulfoxide (DMSO) at the proper concentrations. Human hepatoma HepaRG™ cells were purchased from Thermo Fisher Scientific (Waltham, Massachusetts, USA). For 2 weeks, frozen cells were defrosted and kept alive in Williams’ E medium (Sigma-Aldrich, St. Louis, Missouri, USA) with 10% FetalClone™ II serum (HycloneTM, GE Healthcare, Chicago, Illinois, USA), 1 × L-glutamine, 5 μg/mL human insulin, and 50 μM hydrocortisone hemisuccinate without antibiotics as supplements. To produce differentiated hepatocyte-like features, the medium was then changed to the aforementioned medium plus 2% DMSO for an additional 2 weeks. Cells were grown at 37°C in a humidified 5% CO2 environment. P-nitrophenylphosphate (PNPP) was used in an acid phosphatase (ACP) experiment to measure cell viability (29 (link)).

Accelerated Oxidation Stability Evaluation of Drugs

N-methyl pyrrolidone (NMP) was procured from Sigma Aldrich (Steinheim, Germany). Five of the drug compounds namely, droperidol (DPD), mifepristone (MIF) cimetidine (CIM), naproxen (NAP), clotrimazole (CLO) were purchased from Merck (Sigma Aldrich®, Vienna, Austria), while indomethacin (IMC), nifedipine (NIF), and diclofenac (DIC) were obtained from TCI chemicals® (Tokyo, Japan). Olanzapine (OLA) was purchased from Dr. Reddy’s laboratories (Hyderabad, India). All the selected drugs had a chemical purity above 99.90%, and their chemical structures are shown in Figure 1. Polyvinylpyrrolidone (PVP) K-60 was procured from Merck (Sigma Aldrich®, Vienna, Austria). The weighing of solid powders was done using a laboratory analytical balance. Ultrapure water was obtained from a TKA water purification unit (Vienna, Austria). Accelerated oxidation stability was conducted using a high temperature/pressure oxidation testing device, RapidOxy® (Anton Paar GmbH®, Graz, Austria).
