A competitive immunoassay was used for the determination of F2-isoprostanes in urine (Cayman Chemical, Charlotte, USA). Urine was purified using the solid phase extraction cartridges. The purification and the subsequent ELISA assay were performed following the manufacturer’s recommendations. Plasma protein carbonyls and erythrocyte glutathione were determined spectrophotometrically as described previously [17 (link)]. Oxidation of GSH was prevented using N-ethylmaleimide, which is widely considered the most appropriate blocking agent for preventing glutathione oxidation [18 (link)]. Positive controls have been used in both the ELISA and the spectrophotometric assays.

Hepatic Biomarkers and Cytokines
