Synthesis and Characterization of Polymer-Drug Conjugates

α-Amino-ω-methoxy poly(ethylene glycol) (mPEG-NH2, Mw = 10,000 g mol-1, Mw/Mn =1.05) and Fmoc-NH-PEG-NH2, Mw = 10,000 g mol-1 and mPEG-NH2, Mw = 5,000 g mol−1 were purchased from JenKem Technology (TX, USA). L-leucine and D, L-leucine (Leu), 1, 2-ethylenediamine, 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC), paclitaxel, and other chemicals were purchased from Sigma-Aldrich (St Louis, MO) and were used without further purification. Trastuzumab was obtained from UNMC pharmacy. Fetal bovine serum (FBS), DMEM and RPMI 1640 medium, penicillin, streptomycin, Trypsin–ethylenediaminetetraacetic acid (EDTA) (0.05% trypsin, 5.3 mM EDTA tetra-sodium) and other chemicals were purchased from Invitrogen (Carlsbad, CA, USA). MTT reagent (3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) was purchased from Research Products International (Prospect, IL). Cyanine3 (Cy3) carboxylic acid was purchased from Lumiprobe Life Science Solutions. All other chemicals were of reagent grade and used without further purification.

Niclosamide Formulation Development

Niclosamide (NCL) was purchased from Shanghai Yuanye Bio-Technology Co., Ltd. (Shanghai, China). 1,2-Dioctadecanoyl-sn-glycero-3-phophocholine (DSPC), polysorbate 20 (Tween-20) and polysorbate 80 (Tween-80) were purchased from Tokyo Chemical Industry Co., Ltd. (Tokyo, Japan). L-Leucine (Leu) and methanol were obtained from Sigma–Aldrich (Saint Louis, USA). Acetonitrile (ACN) and formic acid (FA) for HPLC analysis were purchased from Fisher Scientific Co., Ltd. (Pittsburgh, PA, USA) and Aladdin Co., Ltd. (Shanghai, China), respectively. Distilled water (18.2–18.5 MΩ·cm) from a Milli-Q apparatus (Merck, Darmstadt, Germany) was used throughout the experiment.

CCAT2 Silencing and mTOR Agonist Treatment

HGC-27 and SGC-7901 cells were digested with 0.25% trypsin (Gibco), and seeded in 12-well plates at a density of 4 × 105/well. Followed by 24 h of culturing, HGC-27 and SGC-7901 cells were transfected with siRNA-CCAT2 (Generay, Shanghai, China) using lipofectamine 2000 (Thermo Fisher Scientific, USA) according to the manufacturer’s instruction. Cells transfected with siRNA-negative control (siRNA-NC) were considered as the NC, and cells without transfection were considered as the blank control (Control). After the transfection for 48 h, HGC-27 cells in the siRNA-NC and siRNA-CCAT2 groups were treated with 10 mM mTOR agonist, L-Leucine (Leu) (Sigma, USA) for another 48 h.

Schlenk Techniques for Air-Sensitive NCA Synthesis

Schlenk-line techniques were used to handle all compounds, which are sensitive to air and moisture under a nitrogen atmosphere. All chemicals and solvents were ACS reagent grade and used without any purification process unless otherwise noted. Z-L-lysine (ZLL) (Sigma-Aldrich, Schaffhausen, Switzerland), L-leucine (Leu) (Sigma-Aldrich, Tokyo, Japan), L-phenylalanine (Phe) (Sigma-Aldrich, Tokyo, Japan), L-alanine (Ala) (Sigma-Aldrich, Taufkirchen, Germany), and L-valine (Val) (Sigma-Aldrich, St. Louis, MO, USA) N-carboxyanhydrides (NCAs) were prepared based on a procedure reported in the previous papers [28 (link),32 (link)].

Isobutyraldehyde (99%), 2-methylbutanal (95%), phenylacetaldehyde (95%) and methional (98%), 2-methylpentanal (98%), 3-methylpentanal (97%) and O-(2,3,4,5,6-pentafluorobenzyl)hydroxylamine hydrochloride (PFBHA 98%) were supplied by Merck USA. Phenylacetaldehyde-d2 (95%) and methional-d2 were purchased from Eptes (Vevey, Switzerland). Water was purified in a Milli-Q system from Millipore (Bedford, Germany).
