Automated Biochemical Analysis of Murine Urinary Protein

Check if the same lab product or an alternative is used in the 5 most similar protocols
A fully automatic biochemical analyzer was used to determine the urinary protein of mice at 13 weeks old (24 h before treatment) and at 25 weeks old (one day before the end of treatment). Blood was collected from the abdominal aorta and sit at room temperature for 2 h. After centrifuged at 1500 rpm for 10 min, the supernatant was collected. The urine creatinine test kit (Nanjing Jiancheng, C011-1) and urea nitrogen test kit (Nanjing Jiancheng, C013-2) were used to test the specific biofluid of interest, urine, pladma, and serum according to manufacturer’s instructions.

HPLC-based Metabolite Extraction and Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
High performance liquid chromatography (HPLC) grade acetonitrile and methanol were purchased from Thermo Fisher Scientific Co., Ltd. (Fair Lawn, NJ, USA). Ultra-pure water was obtained from Hangzhou Wahaha Group Co., Ltd. (Hangzhou, China), N,O-bis(trimethylsilyl)trifluoro-acetamide (BSTFA) with 1% trimethylchlorosilane (TMCS), O-methoxyamine-HCl (MOX), succinic-d4 acid were purchased from Sigma–Aldrich (NJ, USA). Adenine, 4% tissue cell fixative, yeast powder, urease activity test kit, hematoxylin-eosin staining (HE) staining kit, Masson staining kit and standards for creatinine, uric acid, hypoxanthine, xanthine, urea, etc. were purchased from Beijing Solarbio Science & Technology Co., Ltd. (Beijing, China) and the purity of the above standands are all greater than 98%. The urea nitrogen test kit was purchased from Nanjing Jiancheng Bioengineering Institute (Nanjing, China). Other chromatographic reagents were obtained from domestic reagent companies.

Albumin and Urea Production Kinetics

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 5, 10, 15, and 20 days in the culture systems, culture supernatants were collected, the albumin content was measured using a human albumin ELISA kit (Solarbio, Beijing, China), and the urea content was measured using a urea nitrogen test kit (Nanjing Jiancheng, Nanjing, China). The albumin data are expressed as ng/104 cells·h, and the urea data are expressed as μg/104 cells·h.

Measuring BUN and Serum Creatinine

Check if the same lab product or an alternative is used in the 5 most similar protocols
The blood urea nitrogen (BUN) levels and serum creatinine (Scr) levels were measured using urea nitrogen test kits and creatinine assay kits (Jiancheng Bioengineering Institute, Nanjing, China). The experimental procedures outlined in the instruction manual were followed to determine the blood urea nitrogen and creatinine content.

Biomarkers of Kidney Function

Check if the same lab product or an alternative is used in the 5 most similar protocols
Five mice from each group were killed at the same point by exsanguination under anesthesia before blood samples were collected, followed by centrifuging blood at 3000 rpm for 10 min at 4°C to obtain serum. Urea nitrogen and creatinine were detected using urea nitrogen test kits and creatinine test kits (Nanjing Jiancheng Bioengineering Research Institute, China), respectively. Microalbuminuria was detected using mouse urine microalbuminuria detection kits (Shanghai Enzyme-linked Biotechnology Co., Ltd., China).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
