Inhibition Assay for Cell Motility

Latrunculin A (Sigma–Aldrich), 5‐(N‐Ethyl‐N‐isopropyl) amiloride (EIPA, Sigma–Aldrich), Ouabain (Sigma–Aldrich), Bumetanide (Ro 10–6338, Santa Cruz Biotechnology), Rab7 inhibitior CID 1067700 (Sigma–Aldrich), MyoVin‐1 (Sigma–Aldrich), BAPTA (Sigma–Aldrich), BAPTA‐AM (Sigma–Aldrich), FTY720, 2‐APB and CAI (Tocris Biosciences), Calpain inhibitor I (Millipore Sigma) in dimethylsulfoxide and Gadolinium (III) Chloride (Sigma–Aldrich) in water were used. For all inhibition experiments, cells were incubated in cell culture media with drugs for a duration of 1 h (except Ouabain ≈2 h) and then, high viscosity media was added. Cells were then imaged over a span of 5 h for speed measurements or were incubated over a span of ≈4 h for immunostaining.

Preparation of Uranium(IV) Chloride and Lithium Naphthalenide

(99%, Seulberger) were refluxed and freshly distilled from sodium

Synthesis and Characterization of Multimodal Polymeric Probes

Poly(ethylene glycol) (600)-polybutadiene (1200) copolymer (denoted PEG-PBD) was purchased from Polymer Source (Dorval, Quebec, Canada). Glycol chitosan was purchased from Wako Chemicals (Richmond, VA). DTPA dianhydride, gadolinium (III) chloride, fluorescein isothiocyanate-dextran (FITC-dextran MW 59,000–77,000), tetramethylrhodamine isothiocyanate-Dextran (TRITC-dextran, MW 4400) and poly(ethylene glycol)-block-poly(propylene oxide)-block-poly(ethylene glycol) (PEG-PPO-PEG, MW 4400) were obtained from Sigma-Aldrich. Sulforhodamine B (SRB), tetramethylrhodamine-dextran (TRITC-dextran, 10,000 MW) and tetramethylrhodamine-dextran (TRITC-dextran, 40,000 MW) were obtained from Thermo Fisher Scientific Inc. All other chemicals were used as received. All of the buffer solutions were prepared with deionized water.

Synthetic Interstitial Fluid Protocol

Synthetic interstitial fluid (SIF) comprised (in mM) 107.8 NaCl, 26.2 NaCO3, 9.64 Na-gluconate, 7.6 sucrose, 5.05 glucose, 3.48 KCl, 1.67 NaH2PO4, 1.53 CaCl2 and 0.69 MgSO4, gassed with 95% O2 and 5% CO2 to obtain pH 7.4 [29 (link)]. Stock solutions of the TRPA1 antagonist HC030031, the TRPM8 antagonist AMTB hydrochloride and the TRPV4 antagonist RN 1734 (all purchased from Tocris Biosciences, Ellisville, USA) were made up in absolute methanol. Gadolinium(III) chloride, amiloride hydrochloride hydrate and ruthenium red (all purchased from Sigma-Aldrich, Steinheim, Germany) and the final dilutions were prepared by dissolving the stock solutions in SIF on the day of each experiment. The pH was adjusted to 7.4 by adding drops of HCl or NaOH.

Synthesis and Characterization of CuO/MnO2/Gd2O3 Nanostructures

In this research work, the required chemicals were used as received without further purification. The inorganic salts of transition metals such as copper(II) chloride (CuCl2), manganese(II) chloride (MnCl2), gadolinium(III) chloride, and ammonium hydroxide (NH4OH) were purchased from well-known Sigma-Aldrich, which was deployed to prepare ternary CuO/MnO2/Gd2O3 NSs. As a part of these studies, the analytical grade chemicals 2,4-DNP (2,4-dinitrophenol), 3,4-DAT (3,4-diaminotoluene), pyridine, BH (benzaldehyde), 3-chlorophenol, THF (tetrahydrofuran), methanol, 3-MPHyd (3-methoxyphenylhydrazine), AH (ammonium-hydroxide), coating agent nafion (in 5% ethanol), NaH2PO4, and Na2HPO4 were also bought from the Sigma-Aldrich and deployed as-received. For the details characterization of synthesized CuO/MnO2/Gd2O3 NSs, conventional XRD, XPS, FTIR, UV/Vis and FESEM were implemented to evaluate the structural, crystalline, functional, optical, morphological and elemental analyses. The reliable current versus potential (electrochemical method) was used to determine 3-MPHyd with active CuO/MnO2/Gd2O3 NSs material by using Keithley electrometer (6517A, USA) at room conditions.

Fungal Growth Assay Protocols

Comprehensive Compound Preparation Protocol
