Establishing Diabetic Retinopathy Cell Model

The human retinal pigment epithelial cell lines ARPE-19 were got from American Type Culture Collection (Manassas, VA, USA). The cell culture dishes required here were Dulbecco’s modified Eagle’s medium/nutrient mixture F12 (Procell, Wuhan, China) with 10% fetal bovine serum (Rwdls, Shenzhen, China) and 100 U/mL penicillin/streptomycin. These mediums were placed in a suitable incubator with 95% air and 5% CO2 at 37°C. For the experiments, some of the dishes were added with 30 mM high glucose (HG) for 9 days incubation to establish DR cell model [16 ]. Different concentrations of CAR were added to the established cell model for subsequent experimental studies. The other part was handled only by CAR at the doses of 2.5, 5, 10, 20 μM [15 (link),17 (link)] for 2 h. CAR was supplied by Qilu Pharmaceutical (Jinan, China). Cells were pretreated with 10 μmol/L Nrf2 inhibitor ML385 (MedChemExpress, Shanghai, China) for 24 h and stored for the later experiments.
