Leucine dehydrogenase (LDH) from
Bacillus cereus was in our laboratory. Plasmid pET-28a (+) and
Escherichia coli BL21 (DE3) were procured from Novagen (Nanjing, China) and used as the gene expression vector and host, respectively. Kanamycin, isopropyl-
β-D-thiogalactopyranoside (IPTG), 1-fluoro-2,4-dinitrophenyl-5-L-alanineamide (FDAA), NAD
+, and NADH were obtained from TCI (Shanghai, China). α-keto acids (4-methyl-2-oxopentanoic acid, 3-methyl-2-oxobutanoic acid, 2-oxobutanoic acid, 3,3-dimethyl-2-oxobutanoic acid, 2-oxopropanoic acid, 2-oxo-3-phenylpropanoic acid) and α-amino acids (
L-leucine,
L-valine, L-2-aminobutyric acid, L-
tert-leucine, L-alanine,
L-phenylalanine) were purchased from J&K Chemical (Beijing, China). All other chemicals were purchased from commercial sources, at least reagent grade, and were used without further purification.
