Antimicrobial Activity Evaluation of MRSA and VRSA

Check if the same lab product or an alternative is used in the 5 most similar protocols

S. aureus RN450, ATCC6538, methicillin-resistant S. aureus (MRSA) USA300, and vancomycin-resistant S. aureus (VRSA) Mu50 were wild-type standard strains (Novick, 1967 (link); Garrido et al., 1982 (link); Hiramatsu et al., 1997 (link); Diep et al., 2006 (link)). The other three MRSA isolates, BN226, BN295, and BN508, were clinical wild-type pathogens. All clinical wild-type pathogens were isolated from the wound secretion of inpatients at the First Affiliated Hospital of Anhui Medical University. All isolates (from the Anhui Center for Surveillance of Bacterial Resistance-Huinet, Hefei, China) were grown in tryptic soy broth (TSB) broth medium (Solarbio, Shanghai, China) at 37°C. PEITC was obtained from Sigma-Aldrich (St. Louis, MO, USA). Auranofin was ordered from Yuanye Bio-Technology Co., Ltd. (Shanghai). Other reagents were obtained from Sigma-Aldrich. All prepared solutions were stored at -20°C within 1 month. Wild-type BALB/c mice were purchased from the Experimental Animal Center of Anhui Province (Hefei, China). All experiments involving mice were approved by the Institutional Animal Care and Use Committee of Anhui Medical University (Hefei, China).

Antibiotic Resistance in CRAB Strains

Check if the same lab product or an alternative is used in the 5 most similar protocols
Auranofin (Yuanye, Shanghai, China) and other gold complexes were dissolved in DMF (Sigma Aldrich, USA). All antibiotics (Yuanye, Shanghai, China) were prepared in sterilized Milli-Q water (Millipore, USA). The CRAB strain was isolated from hospital (High-throughput DNA sequencing results are seen in Fig. S5, Tables S1 and S2). The carbapenem-resistant Klebsiella pneumonia (BAA-1705), carbapenem-resistant Pseudomonas aeruginosa (BAA-2108), carbapenem-resistant Escherichia coli (BAA-2140), vancomycin-intermediate Staphylococcus aureus (ATCC 700699), were purchased from the American Type Culture Collection (Manassas, USA). All the strains were grown in Luria-Bertani (LB) broth containing 10 g/L tryptone (Aobox, Beijing, China), 5 g/L yeast extract (AOBOX, Beijing, China) and 10 g/L NaCl (Solarbio, Beijing, China).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
