For isolation of protein in bladder tissue, a commercial kit was used following its protocol (Beyotime). Quantification of Autophagy Markers

Cytosolic and nuclear proteins were extracted from heart tissue samples by using ice-cold radioimmunoprecipitation assay (RIPA) lysis buffer. After protein concentration was measured, equal amounts of cytosolic and nuclear proteins were electrophoresed, blotted, and incubated with antibodies and developed. Equal amount of protein lysates from each heart tissue was added to each well, and β-actin was utilized as a protein marker for normalization.
