Sildenafil Effects on Exercise-Induced Fatigue

Adult male Sprague-Dawley rats (8 weeks old) were used in this experiment. The experimental procedure was performed in accordance with the animal care guidelines of the National Institutes of Health and the Korean Academy of Medical Sciences. The rats were randomly divided into four groups (n=10 in each group): the control group, the exhaustive exercise group, the exhaustive exercise and 20-mg/kg sildenafil citrate-treated group, and the exhaustive exercise and 40-mg/kg sildenafil citrate-treated group. The rats in the sildenafil citrate-treated groups received sildenafil citrate (Sigma Chemical Co., St. Louis, MO, USA) orally once a day for 14 consecutive days at respective dosage. The rats in the control and exercise groups received normal saline orally once a day for 14 consecutive days.

PDE5A1 Enzymatic Activity Assay

PDE5A1 activity was assayed, as described earlier, with minor modification [24 (link)]. Briefly, PDE5A1 (75.3 µg/mL) prepared in Tris–HCl (50 mM; pH = 8.0), MgCl2 (10 mM) and DTT (1 mM) was mixed with 10–30 nM of 3H-cGMP (20,000–30,000 cpm/assay, GE Healthcare, Buckinghamshire, UK) as a substrate as well as pGlu (Sigma-Aldrich, Berlin, Germany; purity ≥99%) and sildenafil citrate (Sigma Aldrich, Berlin, Germany; European Pharmacopoeia (EP) reference standard) as inhibitors (at concentrations ranging from 3 to 10 µM). The enzymatic reaction took place at 25 °C for 20 min and then the reaction was stopped by adding ZnSO4 (0.2 N). Further, the reactant 3H-GMP was accelerated by Ba(OH)2 (0.2 N), while unreacted 3H-cGMP was kept in the supernatant. PerkinElmer 2910 liquid scintillation counter (Thermo Fisher Scientific, Paisley, UK) was utilized to measure the radioactivity of the supernatant through 2.5 ml Ultima Gold liquid scintillation cocktails (Thermo Fisher Scientific, Paisley, UK). The half-maximal inhibitory concentration (IC50) values for test inhibitors were ascertained in the presence of proper concentrations of 3H-cGMP (10–30 nM) and the enzyme (75.3 µg/mL) that hydrolyze up to 70% of the substrate and subsequently calculated by nonlinear regression analysis. Each measurement was conducted in triplicate.

Primary anti-mouse antibodies for GPx and GST were obtained from Santa Cruz Co., USA.

Pharmacological Modulation of Murine Behaviors

Sildenafil Modulates CD56+ Cell Culture

Isolated CD56-positive cells were cultured in 24-well plates (SPL Life Sciences Co., Ltd., Korea), in Opti-MEM Reduced Serum Media (Gibco, Life Technologist) containing 10% FCS (Sigma-Aldrich), 1 U/ml penicillin/streptomycin/100 μg/ml (PAA), 2 mM glutamine (Sigma-Aldrich), 1 mM sodium pyruvate (Fluka), nonessential amino acids (Gibco, Thermo Fisher Scientific), 55 nM 2-mercapthoethanol, 10 ng/ml recombinant human IL-15 (Sigma-Aldrich), 2 ng/ml recombinant human TGF-β-1 (R&D), and 1 μM 5-aza-2′-deoxycytidine (Sigma-Aldrich) in hypoxic (94% N2, 5% CO2, and 1% O2) environment [9 (link)]. The cells were cultured in 1 × 106/ml concentration in two variants: with and without 400 ng/ml of sildenafil citrate (Sigma-Aldrich). The concentration of sildenafil used in the experiments equaled the blood concentration of sildenafil after the oral administration of 200 mg of Viagra in healthy men [35 (link)]. After 7 days of culturing, the cells were harvested for flow cytometry analysis.

Sildenafil Citrate Loaded Nanoparticle Formulation

Quantification of Sildenafil and Metabolites

The following Standards were used in the study: sildenafil citrate (SLD) (Purity > 99%) (Fig. 1) (Sigma-Aldrich, St. Louis, MO, USA), SLD (Viagra) (Pfizer Inc, USA), hydroxyprogesterone (Purity ≥ 99%) (Bayer Pharma AG, Germany) and oestradiol benzoate (Purity ≥ 98%) (Misr Co. for Pharm. Ind., Egypt). Four marketed products (Film coated Tablet, 50 mg) approved in Saudi Arabia were obtained from Drug stores in Riyadh. Chromatographic and analytical grade reagent (AR) were used for extraction and method development. Pre-coated, glass-baked TLC plates obtained from E. Merck (RP-18-60F254; thickness: 0.2 mm; area: 20 × 10 cm) were used for quantification.

Sildenafil and Tadalafil Citrate Quantification
