Immunohistochemical Analysis of Proliferation and Smooth Muscle

Immunohistochemical Analysis of Cancer Markers

The paraffin tissue sections were subjected to dewaxing and rehydration procedures. After performing antigen retrieval, the slides were subjected to blocking using bovine serum albumin (Sango Biotech, Shanghai, China). Immunohistochemical Analysis of Tumor Tissues

Paraffin-embedded tumor tissues from patients and nude mice were sectioned to generate 5 μm sections, which were deparaffinized and rehydrated. After blocking endogenous avidin and biotin, antigen retrieval was performed using sodium citrate buffer at high temperature for 20 min. The antigens were blocked with BSA for 30 min, and sections were incubated with antibodies overnight at 4 °C. The antibodies used were as follows: RACGAP1 polyclonal antibody (Proteintech, cat no. 13739-1-AP, dilution 1:200), MCM3 polyclonal antibody (Proteintech, cat. no. 15597-1-AP, dilution 1:200), PCNA polyclonal antibody (Proteintech, cat. no. 10205-2-AP, dilution 1:100) and MKI67 (Proteintech, cat. no. 27309-1-AP, dilution 1:200). On the following day, the sections were incubated with secondary antibodies, washed three times, stained with DAB for 5 min, counterstained with hematoxylin for 5 min, dehydrated, and sealed with neutral resin. Images were acquired with a digital scanner, and the results were analyzed by ImageJ software.
