Berberine Chloride Treatment Effects

Berberine chloride, ethanol (32221), dimethyl sulphoxide (DMSO)(D2650), and LPS were purchased from Sigma-Aldrich (St. Louis, MO, USA). Anti-CX3CL1 (ab25088), anti-CX3CR1 (ab8021), and anti-ADAM10 (ab1997) antibodies were obtained from Abcam (Cambridge, UK). Anti-phospho-STAT3 (D3A7), anti-RELA (D14E12), and anti-phospho-MAPK14 antibodies were purchased from Cell Signaling Technology (Danvers, MA, USA).
Berberine chloride was dissolved in H2O:ethanol (1:1) for in vivo experiments and in DMSO for in vitro experiments performed in this study.

ADAM10 Protein Expression Analysis

The total proteins were extracted from the treated cells using the RIPA lysis buffer (Beyotime, Shanghai, China), and the concentrations of protein were measured using a BCA Protein Assay Kit (Thermo Fisher Scientific). After separation by electrophoresis on a SDS-polyacrylamide gel, the proteins were transferred to a polyvinylidene difluoride (PVDF) membrane (Millipore, Bedford, MA, USA). Immunoblotting was performed at 4°C overnight with anti-ADAM10 (ab1997; Abcam, Cambridge, UK) and anti-β-actin antibodies (Santa, CA, USA), followed by HRP-linked secondary antibodies. The blots were visualized using an enhanced chemiluminescence (ECL) detection kit (Millipore, Billerica, MA, USA).

Lefkowitz. Immunofluorescence staining was performed with the following primary antibodies: anti-Tuj1 (801201, BioLegend), anti-Sox2 (sc-17320, Santa Cruz), anti-Map2 (Ab5622, Millipore), and anti-GFAP (sc-6170, Santa Cruz).
