Culturing Human Cancer and Normal Cells

Human colon cancer cell lines HCT116 and HT-29, human lung cancer cell lines A549 and H460, and human breast cancer cell line MCF-7 were obtained from the American Type Culture Collection (ATCC, Rockville, MD, USA). Human normal lung epithelial cell line BEAS-2B and human normal colon fibroblast cell line CCD-18Co was obtained from the ATCC. HCT116, A549, H460, and BEAS-2B cells were maintained in RPMI1640 media (Welgene, Daegu, Korea) supplemented with 10% (v/v) fetal bovine serum (FBS) (Hyclone, Rockford, IL) and 1% (v/v) penicillin and streptomycin (Invitrogen, San Diego, CA). HT29 and MCF-7 cells were maintained in DMEM media (Welgene) supplemented with 10% (v/v) FBS and 1% (v/v) penicillin and streptomycin. CCD-18Co cells were maintained in MEM media (Welgene) supplemented with 10% (v/v) FBS and 1% (v/v) penicillin and streptomycin.

Culturing and Characterizing Human CRC Cell Lines

Human CRC cell lines, including HCT116 (a colon carcinoma cell line), HT29 (a colon adenocarcinoma cell line), SW480 (a colon adenocarcinoma cell line), and DLD1 (a colon adenocarcinoma cell line), and the normal colon cell line CCD-18Co were purchased from the Korean Cell Line Bank (Seoul, Republic of Korea) in 2017-2019. Human CRC cells were cultured in Roswell Park Memorial Institute (RPMI) 1640 medium (Welgene, Daegu, Republic of Korea) supplemented with 5% fetal bovine serum (FBS) and 100 U/mL penicillin/streptomycin (Welgene) at 37 °C in humidified incubator with 5% CO2. Patient-derived primary CRC cells and CCD-18Co cells were cultured in Dulbecco's modified Eagle's Minimal Essential Medium (DMEM) (Welgene) supplemented with 10% FBS. Human lung fibroblast cells (MRC5) were a kind gift from Prof. Woo Keun Song (GIST, Gwangju, Republic of Korea) and cultured in Eagle's Minimum Essential Medium (MEM; Welgene) supplemented with 10% FBS. Irradiation was performed using a soft X-ray irradiator (Model M-100, SOFTEX, Tokyo, Japan). All experiments were performed with cells between passages 2-8. All cell lines were routinely tested for mycoplasma contamination every 6 months using the e-MycoTM Mycopasma PCR detection kit (iNtron Biotechnology, Seongnam, Republic of Korea).
