Qingyi Granules Inhibit Liver Fibrosis

Sixty-four male Sprague–Dawley rats weighing 180–220 g were provided by the specific-pathogen free animal center of Dalian Medical University and kept in a clean animal laboratory with ad libitum access to food and water. All animal protocols were approved by the Committee on the Ethics of Animal Experiments of Dalian Medical University (No. AEE19001). Qingyi granules were provided by the Pharmaceutical Department of the First Affiliated Hospital of Dalian Medical University. The herbs in Qingyi granules include Bupleurum chinense, Gardenia jasminoides, Scutellaria baicalensis, Paeonia lactiflora, Corydalis yanhusuo, Saussurea costus, Rheum officinale, and Natrii Sulfas. Each of the voucher specimens was kept in the Clinical Laboratory of Integrative Medicine at the First Affiliated Hospital of Dalian Medical University. The formula preparation, chemical profiling and quality control have been reported by our team previously (Wei et al. 2021 (link)). Emodin (C15H10O5, MW: 270.24, CAS: 518-82-1), an anthraquinone derivative that is also named 2-methyl-4,5,7-trihydroxyanthraquinone and has a light yellow to orange appearance, was obtained from Solarbio Life Sciences Co. (IE0070), China.

High-glucose Dulbecco's modified Eagle's medium was obtained from HyClone Co (Logan, Utah). Fetal bovine serum, trypsin-Ethylene Diamine Tetraacetic Acid (0.05%), and phenol red were provided by Gibco Co, Ltd (Thermo Fisher Scientific, Cleveland, Ohio). Emodin and STC were purchased from Solarbio Science and Technology Co (Beijing, China). PrimeScript RT reagent and SYBR PremixEx Taq II (TliRNaseH Plus) were purchased from TaKaRa Biotechnology Co, Ltd (Dalian, China). RNAex Pro RNA reagent was obtained from Accurate Biology Co, Ltd (Changsha, China). A ROS Detection Assay Kit was purchased from AmyJet Scientific Inc Co, Ltd (Wuhan, China). An α-Amylase (α-AMS) Assay Kit and Gamma-Glutamylcysteine Synthetase (γ-GCS) Assay Kit were provided by the Nanjing Jiancheng Institute of Biotechnology (Nanjing, China). A Micro Reduced Glutathione (GSH) Assay Kit was provided by Solarbio Science and Technology Co (Beijing, China). Rabbit anti-S100A9 was purchased from Cell Signaling Technology, Inc (Beverly, Mass). Rabbit anti-VNN1 was purchased from the Proteintech Group (Chicago, Ill). Goat antirabbit IgG H&L (Horseradish Peroxidase) was obtained from Abcam (Cambridge, UK). An Annexin V-FITC/PI Apoptosis Detection Kit was provided by Dalian Meilun Biotechnology Co, Ltd (Dalian, China).

Emodin Alleviation of High-Fat Diet Induced Obesity in Mice

Healthy C57BL/J mice (male, age 7 weeks, n = 40) were purchased from HFK Bioscience Co., Ltd. (Beijing, China). All mice were housed in the specified pathogens free for 1 week before experiments. Animals were randomly divided into 4 groups according to their diets and treatments: normal chow group (NC, n = 10), HFD group (HFD, n = 10), emodin treatment group (EM, n = 10), and HFD group with emodin (EM-HFD, n = 10). 2 animals in the HFD group and 1 in the HFD-EM group were detected with hyperglycemia. Additionally, 1 mouse in the HFD-EM group died in the intragastric administration. So, we excluded 4 mice in the NC group (2 mice) and EM group (2 mice). After 12 weeks of feeding with normal chow or HFD diet, mice were administrated with emodin (in DMSO) (50 mg/kg) [13 (link)] (cat no. 518-82-1; Solarbio Science & Technology Co., Ltd., Beijing, China) or bacteria-free water of the same frequency and the same volume every other day for 6 weeks (Figure 1(a)). Body weights were measured weekly. All experiments were performed according to the guidance of the Ethics Committee for Experimental Research from the First Affiliated Hospital of Jinzhou Medical University.

SAB4700739) were purchased from Sigma-Aldrich; Merck KGaA. Caerulein was obtained from MedChemExpress. Meanwhile, ARJ21 cells were treated with 20 and 40 µM Emodin, according to previous references (28 (link),29 (link)).
