Cells were harvested at 107 cells/ml by filtration onto 0.45 μm pore size nitrocellulose filters (Whatman), using vacuum, scraped from the membrane and immediately submerged in liquid nitrogen. The frozen cell, was cryogenically pulverized for six cycles of 15 Hz for 30 sec on a Retsch MM301 mixer mill. Sample chambers were pre-chilled in liquid nitrogen and re-chilled between each cycle.

For the filter binding assays, 2 CPS/µL (counts per second) radiolabeled probe was mixed with protein or protein mixture in a 100-µL reaction volume. After incubation for 45 min on ice, the mixture was filtered through Whatman 0.45-μm pore size nitrocellulose filters. Only protein-RNA complexes were retained on the filters and detected by scintillation counting. Data fits were performed with GraphPad Prism 5 using a single site saturation binding mode.
