Transgenic Mice for Immunology Research

C57BL/6 (000664), UBC-GFP (004353), Actb-DsRed (006051), OTI (003831), OTII (004194) and CD45.1 (002014) mice were purchased from the Jackson Laboratories and maintained in our facilities. iFABP-tOVA transgenic mouse line was generously provided by Dr. V. Vezys ("R23">Vezys et al., 2000 (link)). Mice were maintained at the Rockefeller University animal facilities under specific pathogen-free conditions and sentinel mice were tested to be negative for Helicobacter spp. and C. rodentium. Mice were used at 7-15 weeks of age for most experiments. Animal care and experimentation were consistent with NIH guidelines and were approved by the Institutional Animal Care and Use Committee at the Rockefeller University.

Transgenic Mouse Bone Marrow Transfer

Transgenic C57BL/6J mice expressing enhanced GFP (UBC‐GFP) were obtained from Jackson Laboratory (Bar Harbor, ME, USA) Jand used as bone marrow or uterine cell donors. Wild‐type C57BL/6J female mice were obtained from Charles River Laboratories (Wilmington, MA, USA) and used as recipients of bone marrow or uterine cells injection. All animals were maintained in the Animal Facility of Yale University School of Medicine. Mice were housed 4–5 per cage in an animal room exposed to a 12‐hrs light/dark cycle (7:00 a.m.–7:00 p.m.) with food and water provided ad libitum. All animals were treated under an approved Yale University institutional animal care and use committee protocol.

Rosa26confetti (Stock 017492), NOD scid gamma (NSG, Stock 005557), NSG-EGFP (Stock 021937), Rosa26mT/mG (Stock 007676), R26-M2rtTA (Stock 006965), pTRE-H2BGFP (Stock 005104), Prrx1-cre (Stock 005584), Ocn-cre (Stock 019509), Pax7-cre (Stock 010530), Shh-cre (Stock 005622), Mpz-cre (Stock 017927), MIP-GFP (Ins1-EGFP, Stock #: 006864), floxed Stat3 (Stat3fl/fl) mice (Stock 016923) and UBC-GFP (Stock 004353) mice were purchased from Jackson Laboratories. Mfge8−/− (RBRC01726) mice30 (link) were purchased from RIKEN bioresource research center. Shn3 loxp mice31 (link) were reported previously. All mice were maintained on a C57BL6/J background throughout the study except for experiments involving 4T1.2 cells that were conducted in BALB/cJ mice. All animals were maintained in accordance with the NIH Guide for the Care and Use of Laboratory Animals and were handled according to protocols approved by the Weill Cornell Medical College institutional animal care and use committee (IACUC).

Genetically Modified Mouse Strains for Research

C57BL/6J-Ly5.1, OT-I, UBC-GFP and Ai9 mice were obtained from Jackson Laboratories, and strains were maintained in the animal department of The Netherlands Cancer Institute (NKI). Ai9 and OT-I, and UBC-GFP and OT-I mice were crossed to obtain the Ai9;OT-I and GFP;OT-I strains, respectively. Between 5-10 mice, both male and female, of the age of 6 to 15 weeks were used for each experiment. All animal experiments were approved by the Animal Welfare Committee of the NKI, in accordance with national guidelines.

Breeding and Maintaining Transgenic Mice

Experiments were performed with male and female mice (aged 3–5 months), and experimental procedures were approved by the local ethics committee.

C57BL/6N (Charles River), Cdh5-CreERT2 (Tg(Cdh5-Cre/ERT2)1Rha (Wang et al., 2013 (link)), Ubow (Ghigo et al., 2013 (link); Mondor et al., 2016 (link)); Ccl19-Cre (kindly provided by B. Hehlgans)(Wimmer et al., 2012 (link)). Transgenic mice were all backcrossed onto a C57BL/6N background. All mice were bred and maintained in a pathogen-free environment at the animal core facility of the Max-Delbrück-Center for Molecular Medicine Berlin, Germany. Light cycles were at 12 hr intervals, temperature was kept at 22°C, and humidity at 55%, in compliance with the institutional rules. Male and female mice between the age of 8-12 weeks were used in all experiments. Experimental groups were sex-matched according to the origin of the donor Eμ-Myc clone. All experiments were conducted in compliance with the institutional guidelines of the Max-Delbrück-Center for Molecular Medicine and approved by the Landesamt für Gesundheit und Soziales Berlin, Germany (G0104/16; G0052/12; G0373/13; G0058/19; G0044/16).

Transgenic Mouse Models for Hematopoietic Stem Cell Studies

HSC-CreERT/R-DTA mice were generated by crossing R26DTA (Gt(ROSA)26Sortm1(DTA)Lky) and HSC-CreERT (Tg(Tal1-cre/ERT)42–056Jrg)37 (link) and used as recipients (male and female) at the age of 14–16 weeks. All mice were bred and maintained under specific pathogen–free conditions in the animal facility at the Medical Theoretical Center of the University of Technology, Dresden. Experiments were performed in accordance with the German animal welfare legislation and were approved by the “Landesdirektion Sachsen – Referat 24.1”.
