Western Blot Analysis of Mouse Brain Proteins

Mouse brain tissues were lysed in brain tissue lysate (Beijing baiaolaibo Technology) with 1% w/v protein inhibitor (Roche), 1% w/v PhosStop (Roche) for 20 minutes, sonicated for 5 minutes, and centrifuged at 10,000 g for 10 min at 4 C. Protein concentrations were determined using a BCA protein assay kit (Thermo Fisher). The proteins were separated on SDS-PAGE gels and subsequently transferred to a PVDF membrane (Millipore). The membranes were blocked in TBS (Sangon) containing 0.5% milk and 0.5% Tween20 (Sangon) for 1 h at room temperature, incubated overnight with primary antibodies in TBS containing 4% BSA (Sangon), 1% Tween20, then incubated with the appropriate HRP-conjugated secondary antibodies for 2 h at room temperature after extensive washing. The blots were detected using ECL TM Western Blotting Detection Reagents (GE), and acquired images were analyzed with ImageJ software.

Quantum Dot-Antibody Conjugate Biosensor

Cadmium oxide (CdO, 99.99%), zinc oxide (ZnO, 99.99%, power), sulfur (S, 99.98%, power), selenium (Se, 99.99%, power), oleic acid (OA, 90%), 1-octadecene (ODE, 90%), and 2-(N-morpholino) ethanesulfonic acid (MES) were purchased from Aldrich. Mouse anti C-reaction protein monoclonal antibody and CRP antigen were obtained from Abcam (USA). The fluorescence spectra were detected using SpectraMaxi3 (Molecular Devices, Sunnyvale, USA). Images of electrophoresis gels were taken using a gel imaging system (GenoSens1860, Shanghai, China). The sizes of QDs and QD-antibody probe were recorded using dynamic light scattering (Nano-ZS 90, Malvern Instruments, UK). Purified water (18.2 mΩ, Millipore USA) was used in all experiments.

Bacterial Cell Wall Digestion Protocol

Bacteria were fixed in 4% PFA (Beyotime Biotechnology, Cat. P0099) at 4 °C overnight, then washed and permeabilized by 0.04% Tween-20 (Sangon Biotech, Cat. 9005-64-5) in PBS. The cell wall was digested with lysozyme by incubating at 37 °C for 15 min. The lysozyme was ordered from Thermo Fisher (Cat. 90082). Following the cell wall digestion step, bacteria were immediately washed and resuspended in PBS with RNase inhibitor (Thermo Fisher Scientific, Cat. N8080119) for the next step.

Western Blot Protein Analysis

Proteins were separated by 10% SDS-PAGE and then transferred to a polyvinylidene difluoride (PVDF) membrane. After blocking with 5% skim milk in Tris-buffered saline (137 mM NaCl, 20 mM Tris–HCl, pH8.0) containing 0.05% Tween-20 (Sangon Biotech, Shanghai, China), the membrane was incubated with a primary antibody (Abcam, Cambridge, UK) overnight at 4 °C. All reagents were of analytical grade and were used as received without further purification.

Rapid Point-of-Care Microfluidic Biosensor
