Secretion of ET-1, t-PA, and PAI-1 in NG and HG Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Secretion of ET-1, t-PA and PA-1 by NGPC and HGPC exposed to NG, IG or HG medium (48 h) was measured by ELISA. For each ELISA, cell supernatants were prepared by seeding cells in triplicate in 96-well plates (2 × 104 cells/well) and exposing them to NG, IG or HG medium for 48 h under standard conditions. Medium from each triplicate was collected into a single tube and stored at − 80 °C for batch measurement. Samples were processed according to the manufacturer’s instructions for ET-1 (Abcam), t-PA antigen (Zymutest; Hyphen BioMed) and PAI-1 antigen (Zymutest; Hyphen BioMed), with absorbance measured at 450 nm using a Varioskan platereader with with SkanIt Software 2.4.5 (Varioskan Flash, Thermo).

Comprehensive Coagulation Factor Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
FII, FV, FVII, FX (Neoplastine R), FVIII, FIX, FXI, FXII (CK Prest) activity levels were measured by clotting assays using specific factor‐depleted plasma as substrate on Sta‐R (Stago, Paris, France). Fibrinogen level (Clauss method, Thrombin Reagent; Siemens, Marburg, Germany), FXIII activity and vWF antigen and activity (FXIIIact/subs, vWF Reag and vWF Ac Reagens; Siemens) measurements were performed on a Sysmex CS2100i. α2‐antiplasmin level was measured using a chromogenic assay (Stachrom; Stago), tissue plasminogen activator (tPA) and plasminogen activator inhibitor 1 (PAI‐1) activity were measured using bio‐immunoassays (Zymutest, Hyphen‐BioMed, Neuville‐sur‐Oise, France).

The Bio-Plex ProTM human cytokine 27-plex assay (Bio-Rad, Vienna, Austria) was used to quantify interleukin (IL)-1 beta, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-12p70, IL-13, IL-15, IL-17A, and IL-1 receptor antagonist (IL-1ra); basic fibroblast growth factor (bFGF); granulocyte colony-stimulating factor (G-CSF); granulocyte-macrophage colony-stimulating factor (GM-CSF); interferon-gamma (IFN-γ); the chemokines CXCL10 (IP-10, interferon-inducible protein 10), CCL2 (MCP-1, monocyte chemotactic protein-1), CCL3 and CCL4 (MIP-1α and MIP-1β, macrophage inflammatory protein-1 alpha and beta); CCL5 (RANTES, regulated on activation, normal T-cell expressed and secreted); CCL11 (eotaxin, eosinophil chemotactic protein); platelet-derived growth factor (PDGF); tumor necrosis factor-alpha (TNF-α); as well as vascular endothelial growth factor (VEGF). Selected plasma samples were further characterized with a membrane-based antibody array (Proteome ProfilerTM ARY002, R&D Systems, Minneapolis, USA) detecting 102 cytokines, chemokines, and growth factors. Plasminogen activator inhibitor-1 (PAI-1) and human endothelial cell-specific molecule-1 (ESM-1, endocan) were quantified by enzyme-linked immunosorbent assays (Zymutest, Hyphen BioMed, Neuville-sur-Oise, France, and Lunginnov, Lille, France, respectively).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
