To detect CD133 and EpCAM double positive CSC, the isolated mononuclear cells were incubated with FcR blocking reagents, and the mixture was stained using anti-EpCAM FITC (BD biosciences, San Jose, CA, USA) and anti-CD133/1(AC133) PE (Miltenyi Biotec GmbH, Bergisch Gladbach, Germany) antibodies for 30 minutes at 4℃. The expressions of CD133 and EpCAM were quantitatively measured by FACS Calibur (BD biosciences) and analyzed by CellQuest software (BD biosciences).

Matched subpopulations were separated as described previously [39] (link), [40] (link). In brief, cells were stained at a concentration of 107 cells per 100 µl of buffer. Anti-CD133/1(AC133)-PE (MiltenyiBiotec) antibody was used for flow cytometric sorting/assay. For all experiments, samples were sorted on a BD FACS Aria II and analyzed on a BD LSR II flow cytometer using BD FACS Diva Software (BD Bioscience). Side scatter and forward scatter profiles were used to eliminate debris and cell doublets.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
