Quantifying miR-30d-5p and GLDC Expression

Total RNA was extracted using TRIzol reagent (Invitrogen, CA) according to the manufacturer’s instructions. Reverse transcription reactions were performed with 1 μg of total RNA using FastQuant RT kit (TIANGEN Biotech, Beijing, China). The sequence of the miR-30d-5p stem loop is 5ʹ-GTCGTATCCAGTGCAGGGTCCGAGGTATTCGCACTGGATACGACCTTCCA-3ʹ. GLDC expression was quantified using a SYBR qPCR Kit (TIANGEN Biotech) according to the manufacturer’s instructions. All samples were run in triplicate. The endogenous RNA reference gene used was 18s ribosomal RNA. The relative expression levels were evaluated using the 2−ΔΔCt method20 (link). Primers are listed in Supplementary Table S1.

RNA Extraction and qPCR Analysis of DDX27 and MVP

Complementary single-strand DNA (cDNA) was transcripted using FastQuant RT kit (With gDNA, TIANGEN, Beijing, China). The expression levels of DDX27 and MVP were determined by qPCR using SYBR qPCR Kit (TIANGEN, Beijing, China). The primer sequences are listed in Table S1.
