Microencapsulated BMSC Histology in Bovine Discs

Healthy, unprimed and primed microencapsulated-BMSC-treated bovine discs were stained for cell morphology after 28 d of whole organ culture. Briefly, slides were permeabilised using 0.5 % Triton X-100 and stained with rhodamine phalloidin at 200 U/ mL (diluted 1 : 40; Sigma-Aldrich) to stain the actin cytoskeleton. 4′,6′-diamidino-2-phenylindole (DAPI, diluted 1 : 500; Sigma-Aldrich) was used to stain the nucleus and rinsed with PBS. Subsequently, stained slides were imaged with a Leica SP8 scanning confocal at 358/524 and 540/565 nm wave-length (40× magnification) and assessed using the Leica Application Suit X (LAS X) software.
