Quantifying Bacterial DNA in Mosquitoes

The amount of total bacterial DNA was determined in triplicate for individual mosquitoes using QuantiTect SYBR Green qPCR (Qiagen) with the same primers utilized for 16S sequencing (V3V4 hypervariable region). A standard curve was generated by extracting DH5-Alpha Escherichia coli, quantitating DNA utilizing a NanoDrop 2000 and calculating genome copies. A 10-fold serial dilution was created from 102 to 108 genome copies.
