Quantification of Antibiotics and Pesticides

Antibiotics and pesticide residues were analyzed using analytic grade reagents. Standards of high purity grades (>99%) for 26 antibiotics belonging to four families: Sulfonamides (sulfacetamide, sulfisomidine, SMT, sulfapyridine, sulfamerazine, sulfameter, sulfachloropyridazine, sulfamethoxazole, sulfabenzamide, sulfadimethoxine, sulfaquinoxaline, sulfamethoxypyridazine, and sulfadoxine), quinolones (nalidixic acid, oxolinic acid, CIP, norfloxacin, ofloxacin, flumequine, danofloxacin, enrofloxacin, lomefloxacin, and sarafloxacin), tetracyclines (tetracycline and oxytetracycline), and β-lactams (ampicillin) in addition to a hundred pesticide standards were procured from Sigma-Aldrich. Each pesticide stock solution was accurately prepared in acetonitrile at a concentration of 1 g/L. The working standards were freshly prepared by diluting the stock solution in acetonitrile to the desired concentration. Both stock and working solutions were stored at −20°C.
High-performance liquid chromatography (HPLC)-grade water, HPLC-grade acetonitrile, disodium ethylenediaminetetraacetate dihydrate (Na2 EDTA), and magnesium sulfate (MgSO4) were also supplied by Sigma-Aldrich.

Reagents for Cell Death Assays

Necrostatin-1 was purchased from FOCUS Biomolecules, and 2-mercaptoethanol was purchased from MP Biomedicals. z-VAD-fmk was purchased from Peptide institute. Trolox, sulfadiazine, sulfadimethoxine, sulfadoxin, and sulfamethoxypyridazine were purchased from Tokyo Chemical Industry. Cl-amidine was purchased from Cayman chemicals. Ionomycin was purchased from Merck Millipore.

Analytical Method for Sulfonamide Antibiotics

Ammonium molybdate tetrahydrate (H24Mo7N6O24·4H2O) (99%), thioacetamide (TAA) (98%), sulfathiazole (STZ), sulfadimidine (SDD), sulfadiazine (SDZ), sulfamethoxazole (SMZ), sulfacetamide (ST), sulfachloropyridazine (SCD), N4-phthalylsulfathiazole (PST), succinylsulfathiazole (SST), sodium hydrogen phosphate (Na2HPO4), sodium dihydrogen phosphate (NaH2PO4), methanol for HPLC (99.9%) and acetonitrile for HPLC (99.9%) were purchased from Sigma-Aldrich (St Louis, USA). Acetone, sodium hydroxide (NaOH) and hydrochloric acid (HCl) were purchased from Beijing Chemical Works (Beijing, China). Polyethylene glycol (PEG4000) was purchased from Merck (Germany). Ultrapure water (18.2 MΩ cm) was prepared using a Milli-Q Gradient ultrapure water system (Millipore, Milford, MA, USA).
The eight SAs were dissolved in a 0.1 mol l−1 NaOH solution to prepare standard stock solutions of 2.0 mg ml−1 for each SA, and these standard solutions were stored at 4°C and protected from light. The working solutions to be analysed were diluted with ultrapure water. The calibration standards of SAs were prepared with five levels of concentration in the range of 0.3 to 30 µg ml−1.

Analytical-Grade Chemicals in Study

The chemicals used are Cu(NO 3 ) Aesar), sulfanilamide (Sigma-Aldrich), sulfacetamide (Sigma-Aldrich), sulfathiazole (Sigma-Aldrich), sulfamethoxazole (Sigma-Aldrich), benzotriazole (Alfa Aesar) and bisphenol A (Merck). All the experiments were conducted using the deionized (DI) water (18.2 MΩ cm).
