1

Comprehensive Hematological and Biochemical Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Retro-orbital or tail vein blood was collected into EDTA-coated tubes for hematology or heparinized tubes for plasma preparation. Complete blood counts were determined using a Scil Vetabc system. Plasma was analyzed for bilirubin, iron, and ferritin using an Olympus AU400 analyzer (Beckman Coulter) with reagents supplied by Beckman Coulter or Randox. Erythropoietin was determined using a Meso Scale Discovery array.

Plasma Biomarker Measurement Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected under terminal anesthesia into either EDTA-coated tubes or heparinized tubes for plasma or serum preparation. Clinical chemistry analysis of plasma was performed with the Olympus AU400 analyzer (Beckman Coulter Ltd., High Wycombe, United Kingdom). Concentrations of Cbl in plasma were measured with the ADVIA Centaur immunoassay analyzer (Siemens).

Plasma Clinical Chemistry Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected under terminal anaesthesia into heparinized tubes for plasma preparation. Within 1 hour of collection, blood samples were centrifuged and plasma recovered and stored at -20°C until analysis. Clinical chemistry analysis of plasma was performed using the Olympus AU400 analyzer (Beckman Coulter Ltd) and was blinded to the operator via barcodes.
The majority of parameters were measured using kits and controls supplied by Beckman Coulter. Samples were also tested for haemolysis. Four parameters were measured by kits not supplied by Beckman Coulter: transferrin, ferritin (Randox Laboratories Ltd), fructosamine (Roche Diagnostic) and thyroxine (Thermo Fisher).

Serum Transaminase Assay in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
A serum alanine aminotransferase (ALT) Beckman Coulter test kit (OSR 6107) and aspartate aminotransferase (AST) were performed on the sera harvested at sacrifice from experimental mice according to the manufacturer’s protocol and analyzed using a Beckman Coulter AU400 analyzer by the Clinical Biochemistry Unit, Women’s Hospital, Edgbaston, Birmingham, U.K.

Comprehensive Blood Profiling in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
At 16 weeks, blood was collected by puncture of the retro-orbital sinus under terminal anaesthesia within 1−3 hours of lights on and collected into ethylenediaminetetraacetic acid-coated tubes (Kabe Labortechnik, Numbrecht, Germany) for hematology (Scil Vetabc, Montpellier, France) and into heparinized tubes (Kabe Labortechnik) for plasma preparation. A total of 26 parameters were determined from plasma using an Olympus AU400 analyzer (Beckman Coulter, High Wycombe, UK). Insulin and erythropoietin were determined using a Meso Scale Discovery array (Rockville, MD) and interleukin 6 was measured by enzyme-linked immunosorbent assay (eBioscience, Hatfield, UK).

Standardized Blood Sample Collection Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Fasting blood samples were collected by qualified nurses in the Clinical Laboratory unit at the Department of Sport and Health. Spanish Agency for Health Protection in Sport (AEPSAD). Complete blood counts were determined from VacutainerTM EDTA tubes (Becton Dickinson, Franklin Lakes, NY, USA) by standardized clinical laboratory procedures using an automated hematology analyzer (ADVIA 120; Siemens Healthcare Diagnostics Inc., Deerfield, IL, USA) within 4h after extraction. For serum separation, blood samples were collected in VacutainerTM gel tubes (No additive (Z) [40 ,41 (link),42 (link)] (Becton Dickinson, Franklin Lakes, NY, USA). Samples were stored at room temperature for at least 15 min to allow clot formation and then immediately centrifuged for 15 min at 4000 rpm and 10 °C. Serum samples were aliquoted and analyzed to assess basic biochemistry in an AU400 analyzer (Beckman Coulter Inc., Brea, CA, USA) and stored at −80 °C for later analysis.

Determination of Plasma Biochemical Parameters

Check if the same lab product or an alternative is used in the 5 most similar protocols
Determination of standard biochemical parameters in plasma -glucose, triglycerides, cholesterol, uric acid, alanine aminotransferase (ALT), aspartate aminotransferase (AST) and C-reactive protein (CRP)was carried out using the appropriate kits from Beckman Coulter on AU 400 analyzer (Beckman Coulter). All measurements were performed and evaluated by following the manufacturer's instructions. Thiobarbituric acid reactive substances (TBARS) were determined after extraction with n-butanol by spectrofluorimetric method (excitation wavelength 528 nm, emission wavelength 558 nm, spectrofluorometer Cary Eclipse, Varian) (Solichová et al. 2003) .
For determination of GSH/GSSG ratio, ragent kit BIOXYTECH® GSH/GSSG-412 TM (OxisResearch, a division of OXIS Health Products, Inc.) was used; absorbance was read on spectrophotometer ECOM 6122 (Eppendorf).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
