RPMI 1640 (Gibco, Life Technologies) complete medium supplemented with 10% fetal calf serum (FCS), 100 units/ml penicillin, 100 μg/ml streptomycin (all from GIBCOBRL®, Life Technologies™), 1 M HEPES buffer solution (Gibco, Life Technologies), 100 mM Sodium-pyruvate (Gibco, Life Technologies), MEM NEAA (100x) (Gibco, Life Technologies), 2-Mercaptoethanol (Sigma), Mycobacterial antigens used include: PPD for in vitro use (Statens Serum Institute, Copenhagen, Denmark) and ds-Red fluorescent protein expressing BCG (strain Myc3305, provided by Dr. Brigitte Gicquel) (Abadie et al., 2005 (link)).

Multi-Marker Immunophenotyping of T Cells

Anti-CD3-FITC (Clone UCHT-1), anti-CD4-APC (Clone MEM-241), anti-CD8-APC (Clone UCHT-4), and anti-CD62L-APC (Clone LT-TD180) were purchased from Immunotools. Anti-CD45RA-PE-cy7 (Clone HI100) was purchased from Invitrogen (Carlsbad, CA, USA). Freshly isolated PBMCs (1×106 cells) and the transduced T cells (1×105 cells) were incubated with specific antibodies (1:100) at 4°C for 45 minutes, and then examined by FACSVerse™ flow cytometer (BD Biosciences). The acquired data was analyzed using FlowJo software.
