Quantification of Glucosinolates in Brassicas

Chemicals and reagents were obtained from the following commercial sources (purity in parenthesis): Gluconapin (3but) (84%), glucoerucin (4mtb) (90%), glucobrassicin (I3M) (86%), glucoraphanin (4msb) (89%), glucoiberin (3msp) (98%) and internal standard, sinalbin (99%) were purchased from PhytoLab GmbH & Co. KG (Vestenbergsgreuth, Germany). 4-Hydroxyglucobrassicin (4OHI3M) (95%), 4-methoxyglucobrassicin (4MOI3M) (94%) and neoglucobrassicin (NMOI3M) (97%) were acquired from Phytoplan, Diehm & Neuberger GmbH (Heidelberg, Germany). Acetonitrile and methanol (LC/MS grade solvents) were obtained from Fisher Chemical (Roskilde, Denmark). Water was obtained from a MilliQ purifier (Millipore, MA, USA). Glacial acetic acid was purchased from Merck (Darmstadt, Germany).

Glucosinolate Determination in Root Powder

For glucosinolate determination around 25 mg of frozen root powder were extracted by adding 1 mL of MeOH:water (70:30). The mixtures were homogenized in a Tissue Lyser (Retsch MM 400, Haan, Germany) and incubated for 15 min at 80 °C to inactivate myrosinase. Then, homogenates were centrifuged for 20 min at 16,000g. Glucosinolates were determined from supernatants by ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC/Q-TOF-MS) analyses using an Acquity UPLC from Waters (Milford, MA, USA) interfaced to a Synapt G2 QTOF from Waters (Milford, MA, USA)with electrospray ionization as described previously [77 (link)]. Quantitative Analysis of Glucosinolates

High-purity (>98%) GSL standards were purchased from PhytoLab GmbH & Co. KG (Vestenbergsgreuth, Middle Franconia, Germany): sinigrin (SIN), gluconapin (NAP), glucobrassicanapin (GBN), progoitrin (PRO), glucoerucin (ERU), glucoraphenin (RAE), glucoraphanin (RAA), glucobrassicin (GBC), glucotropaeolin (TRO) and gluconasturtiin (NAS). 4-Methoxyglucobrassicin (4ME) and neoglucobrassicin (NEO) were purchased from Extrasynthese (Z.I Lyon Nord, Genay Cedex France) and glucoalyssin (ALY) was purchased from Shanghai ZZBIO Co., Ltd. (Shanghai, China). HPLC-grade methanol, LC/MS-grade methanol and LC/MS-grade formic acid were supplied by Fisher Scientific (Pittsburgh, PA, USA). Ultrapure water was obtained using Milli-Q purification systems (Millipore, MA, USA). Individual stock solutions of the 13 GSLs were prepared in 70% methanol.

Glucosinolate Quantification Protocols
