Alkaline Phosphatase Activity in C3H10T1/2 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
C3H10T1/2 cells were seeded in 96-well culture plates (1 × 104 cells/plate) and cultured in 0.2 ml of a DMEM/F12k medium (HyClone, USA) supplemented with 10 % fetal calf serum (FCS, Gibco, USA) and 4 μl of protein extracts that were obtained after 2, 4, 6, and 8 weeks. Medium without fetal bovine serum was used as the control. Alkaline phosphatase (ALP) activity was determined after 1, 3, 6, and 9 days using an ALP ELISA kit (Jiancheng, Nanjing, China). Enzyme activity is expressed as absorbance per microgram of protein per minute (μg/min/μgprot).

Alkaline Phosphatase Activity Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells cultured for 14 days were assessed for ALP activity. ALP staining was performed using the nitroblue tetrazolium (NBT) and 5-bromo-4chloro-3-indolyl phosphate (BCIP) (NBT/BCIP) staining kit (C3206; Beyotime) according to the manufacturer's instructions. And ALP quantification was performed using the ALP ELISA kit (A059-2; Nanjing Jiancheng Bioengineering Institute).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
