Quantitative Analysis of Glycogen in Sepsis

PAS staining was used to assess the glycogen content in the kidneys and liver of sepsis mice. The procedure of PAS was carried out as described previously [19 (link)]. Briefly, the kidney and liver tissue sections were deparaffinized in xylene, hydrated in graded ethanol, and then fixed in methyl Carnoy’s solution, followed by washing 3 times with phosphate-buffered saline (PBS). Sections were then incubated with 0.5% periodic acid, washed in PBS, and stained with Schiff’s reagent. Images were acquired using an Eclipse 90i microscope at 200× magnification (Nikon, Tokyo, Japan) equipped with a DP71 CCD camera image capture system (Olympus, Tokyo, Japan). The images were analyzed using Image-Pro Plus version 6.1, and glycogen content of each tissue is expressed as a percentage of the area that stained positive glycogen (% Area PAS+).

Histopathological Evaluation of Lung Tissues in PSP

All standard hematoxylin and eosin (H&E) stained histologic sections from lung tissues of PSP patients were reviewed using light microscopy. Each tissue slide was evaluated in obvious histologic lesions and was read by the same specialist in pulmonary pathology. Pathological findings including lung tissue features (bleb/bulla, fibrosis, pathologic hyperplasia, hemorrhage, granulation tissue, and cholesterol cleft) and cell features (macrophage, giant cells, eosinophil, lymphocyte, histiocyte, and neutrophil) were recorded. Tissue pathologic images were taken and analyzed using the Olympus fluorescent microscope (Tokyo, Japan) equipped with DP71 CCD camera image capture system.
