Culturing and Inoculating BT-474 Breast Cancer Cells

BT-474 human breast cancer cells were purchased from the American Type Culture Collection, authenticated and tested to be negative for common mouse pathogens and for mycoplasma. The cells were cultured in DMEM/F-12 (Sigma-Aldrich) supplemented with 10% heat-inactivated fetal bovine serum (iFBS, Sigma-Aldrich) in a humidified incubator at +37°C and 5% CO2. 1 × 106 of BT-474 cells suspended in 20 μl of 1x phosphate-buffered saline (PBS) was used per inoculation to the mice. Before and after the inoculation, cell viability was determined (NucleoCounter®, NC-200, ChemoMetec) and it was above 80%.

Cultivation of Human Breast Cancer Cells
