Fluorescent PEG Conjugation Protocol

To a reaction vial equipped with a stir bar was added 8 arm PEG DBCO (Mn = 20 kg mol−1, 2.4 g, 0.12 mmol) and PBS (19 mL). Fluorescein-azide (Lumiprobe (Cockeysville, MD), FAM azide, 6-isomer) stock solution was made in DMSO (10 mg/mL). FAM azide (1 mL, 0.015 mmol) was added to the reaction vial and stirred for 1 h at room temperature in the dark. The functionalized PEG DBCO was then dialyzed extensively against water (MWCO 8 kDa) protected from light, then lyophilized.
