Plasma Isolation and PBMC Cryopreservation

Whole blood was spun at 200×g for 10 min to separate plasma from cells. Plasma was then centrifuged at 1000×g for 10 min to remove platelets and the supernatant was stored at −80 °C till needed for antibody assays. PBMCs were isolated on a Ficoll-Paque density gradient medium (Histopaque-1077, Sigma), washed with 1X PBS, and resuspended in 90 % FBS/10 % DMSO (D2650, Sigma Aldrich USA). Cells were stored in liquid nitrogen in aliquots of 5–10 million per vial and thawed as needed for experiments.
