Human mesenchymal stromal cells were seeded in a 24-well plate at approximately 60~70% confluence. After 24 h, each cell was treated with naked oAd/RLX or oAd/RLX-PCDP complex at various MOIs for 18h. Then, the infected cells were washed twice with PBS and incubated with DMEM (5% FBS) for 4 and 8 days. Then, both supernatant and cells were collected, and the mixture underwent three cycles of freezing and thawing. The copy number of Ad genomes was analyzed by real-time quantitative PCR (Taqman PCR detection; Applied Biosystems, Foster City, CA) as previously described [40 (link)]. Samples were amplified for 40 cycles in an ABI 7500 sequence detection system (Applied Biosystems) and analyzed in triplicate and data were processed by the SDS 19.1 software package (Applied Biosystems).
