Phospho p70s6k pp70s6k

Manufactured by Cell Signaling Technology
Sourced in United States

Phospho-p70S6K (pp70S6K) is a monoclonal antibody that detects the phosphorylated form of the p70 ribosomal protein S6 kinase (p70S6K). p70S6K is a serine/threonine protein kinase that plays a crucial role in the regulation of cell growth, proliferation, and protein synthesis in response to various extracellular signals.

Proteins were loaded in equal amounts and separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis and transferred onto polyvinylidene difluoride membranes (Merck Millipore). Membranes were incubated with the primary antibodies directed against the following molecules: α-SMA (Sigma-Aldrich) PDGF receptor-β (Cell Signaling Technology, Danvers, MA), collagen 1α1 (R&D Systems, Minneapolis, MN), JNK, phospho-JNK (pJNK), ERK1/2, phospho-ERK1/2 (pERK), SMAD2, phospho-SMAD2 (pSMAD2), AKT, phospho-AKT (pAKT), p70S6K, phospho-p70S6K (pp70S6K) (all from Cell Signaling Technology), GAPDH (Abcam, Cambridge, United Kingdom) and β-actin (Sigma-Aldrich). The following secondary antibodies were used as needed: a goat-anti-mouse-IgG-HRP antibody (Bio-Rad, Feldkirchen, Germany), a goat-anti-rabbit-IgG-HRP (Bio-Rad), or a goat-anti-sheep-IgG-HRP antibody (R&D Systems). Visualization was performed using the ChemoCam (INTAS, Homburg, Germany) after incubation with Clarity Western ECL Substrate (Bio-Rad).

Rg1 (purity > 99%) was kindly provided by Professor Kim, Si-Kwan (Konkuk University, Chungju, Korea). Fetal bovine serum (FBS), horse serum (HS), and Dulbecco modified Eagle's medium (DMEM) were obtained from Gibco (Grand Island, NY, USA). Antibodies were as following: MHC (myosin heavy chain; the Developmental Studies Hybridoma Bank, Iowa, IA, USA), Akt, phospho-Akt (pAkt), p70S6K, phospho-p70S6K (pp70S6K), mTOR, phospho-mTOR (pmTOR), phospho-p38MAPK (pp38) (Cell Signaling Technology, Beverly, MA, USA), pan-Cadherin (Sigma, St. Louis, MO, USA), p38MAPK, MyoD, Myogenin, E2A, and α-Tubulin (Santa Cruz Biotechnology, Santa Cruz, CA, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
