Rabbit anti-flag, mouse anti-GFP, mouse anti-α-tubulin and rabbit anti-caveolin-1 antibodies were purchased from Cell Signaling. Rabbit anti-TfR antibody was a Proteintech Group product. The rabbit polyclonal antibodies for GM130, EEA1 and LAMP1 were all ABclone products. Immunostaining of GalNAc-T14 and TGN46

The suspension of Dakiki cells containing 2 × 105 cells27 (link) was pelleted onto a microscopic slide using a CytoCentric 350 (Domel, Otoki, Slovenia). Cells were fixed using 4% paraformaldehyde in PBS, permeabilized with 0.1% Triton X-100 in PBS, blocked with 10% FBS in PBS for 1 hour, and stained with mouse anti-GalNAc-T14 mAb (1:100, Santa Cruz Biotechnology) and rabbit anti-TGN46 (marker of trans-Golgi network, 1:200, Abcam) antibodies in 10% FBS in PBS overnight at 4 °C. After washing, goat anti-mouse IgG-FITC (1:1,000, Sigma-Aldrich; Merck) and goat anti-rabbit IgG-Cy3 (1:2,000, Invitrogen) secondary antibodies in the 10% FBS in PBS were added and incubated for 1.5 hours at room temperature. Cover slips were mounted using Vectashield medium with DAPI (Vector Laboratories, Inc., Burlingame, CA) for fluorescence microscopy (Leica Microsystems GmbH, Wetzlar, Germany).
