FITC-dextran Oral Gavage in Mice

Mice were pre-bled to determine background signal following by OG with 10 mg FITC-dextran (4kDa, Sigma) in 0.2 ml PBS. Total serum was isolated 4 h later by cardiac puncture. Serum was diluted 1:5 in PBS and fluorescence was measured with excitation and emission wavelengths of 485 and 538, respectively, using a SpectraMax M2 microplate reader (Molecular Devices). Fluorescence values were quantified using a standard curve. As a positive control, WT mice were given 3% dextran sulfate sodium salt (36–50 kDa, MP Bio) ad libitum in their drinking water for 7 d prior to FITC-dextran gavage.

Bachu Mushroom Bioactive Compounds

Detailed Reagent Specifications for Colitis Study

Reagent grade Dextran Sulfate Sodium salt (DSS, reagent grade, M.W 36–50kDa, Ref = 160110) was purchased from MP Biomedicals (Solon, OH). Duoset ELISA kits for mouse and human Lcn2 and keratinocyte-derived chemokine (KC) were obtained from R&D Systems (Minneapolis, MN). α-Hu-Lcn2 SYBR® Green mix and qScript cDNA synthesis kit was procured from Quanta Biosciences (Gaithersburg, MD). Guaiacol (2-methoxyphenol) was obtained from Alfa Aesar (Ward Hill, MA). Myeloperoxidase (MPO) was procured from Sigma (St. Louis, MO). Ly6G antibody was purchased from Abcam (Cambridge, MA). Rat anti-mouse IL-10 receptor (IgG1) monoclonal antibody (αIL-10R) and rat anti-mouse PD-1 monoclonal antibody (IgG1 isotype control) were procured from BioXcell (West Lebanon, NH). Anti-mouse CD45-Alexafluor 594 and CD326 monoclonal antibodies were procured from BioLegend (San Diego, CA) RegIIIγ and Ang4 antibodies are kind gift from Dr. Lora Hooper, UT Southwestern Medical Center, TX. All other fine chemicals used in present study were reagent grade and procured from Sigma.

Chitosan-based Nanocarrier Formulation

Ultrapure CS hydrochloride salt (Protasan UP CL 113, Mw 125 kDa, deacetylation degree of 86%) was purchased from Novamatrix (Sandvika, Norway). Dextran sulfate sodium salt (Mw 6–8 kDa) was bought to MP Biomedicals (Illkirch, France). Sodium hyaluronate (HA; Mw 57 kDa) was purchased from Lifecore Biomedical (Chaska, MN). Poly‐l‐glutamic acid sodium salt (Mw 15–50 kDa), and chondroitin‐6‐sulphate sodium salt were obtained from Sigma‐Aldrich.

Colitis-Associated Tumor Development Protocol

The AOM/DSS model was performed to study colitis-associated tumor development with Azoxymethane (Sigma-Aldrich, Saint Louis, MO, USA) and Dextran sulfate sodium salt (MP Biochemicals, Illkirch, France), as described previously [36 (link)]. When using tamoxifen inducible strains, tamoxifen (Cayman Chemical, Ann Arbor, MI, USA) was dissolved in sunflower oil and injected intraperitoneally for five consecutive days one week before each DSS treatment. Each of the three DSS cycles lasted one week and was followed by two weeks of water. tamoxifen 1 mg was applied in a total volume of 100 µL per injection, and all experimental controls obtained the same treatment.

ROS Assay Kit was from Beyotime (Nanjing, China). All other chemicals were of general reagent grade (unless stated).
