Multiplex Cytokine Quantification in Mice

Chemokines and cytokines were quantified by a Cytokines Mouse Magnetic 20-Plex Panel for Luminex (Life Technologies), and a ProcartaPlex mouse IFN-α/IFN-β panel (eBioscience) according to the manufacturer's instructions and data were acquired using a Bio-Plex 200 system (Bio-Rad Laboratories, UK). The concentration of cytokines in each sample was determined according to the standard curve using the Bio-Plex 6 software (Bio-Rad Laboratories). The concentration of CXCL10, CCL2, and TNF-α was additionally measured using mouse DuoSet ELISA (R&D) according to the manufacturer's instructions. IL-6 was detected by ELISA using MP5-20F3 capture antibody and biotinylated MP5-32C11 detection antibody (both from BD Pharmingen). Data were acquired on a SpectraMax Plus plate reader (Molecular Devices) and analysed using SoftMax software (version 5.2).

Luminex-based Cytokine Quantification

Multiplex Cytokine Profiling in Plasma

The analysis was performed according to the manufacturer’s instructions. A Bio-Plex 200 instrument (Bio-Rad) and its software (Bio-Plex 6.1) were used for analysis.

Cytokine Profiling via Luminex Assay

Cell-culture supernatants were analyzed for the presence of IL-17A, IFN-g, and TNF-a by using Bio-Plex Pro Human Th17 Cytokine Assays (Bio-Rad Laboratories), according to the manufacturer's instructions. Multiplex Cytokine Analysis of Serum Samples

Whole blood was collected by venipuncture at the enrollment visit. Blood for cytokine analysis was collected into serum separator tubes and centrifuged and aliquoted into cryovials following a standard protocol during or immediately following the study visit. Serum aliquots were frozen and stored at -80°C pending shipment to the biorepository and subsequently to the analyzing laboratory. As freeze-thaw cycles can alter cytokine measures, samples in our analysis were not freeze-thawed[18 (link)]. Total numbers of eosinophils and neutrophils were determined by the product of the differential and total cell count. Using 25 μL of participant serum, cytokines were quantified according to the manufacturer’s instructions using a premixed 38 Plex MILLIPLEX MAP Human Cytokine/Chemokine Magnetic Bead Panel (Millipore Corporation, Billerica, MA) and analyzed on a Luminex 100/200 analyzer (BioRad, Hercules, CA). Serum cytokine concentrations were determined using Bio-plex 6.1 software (BioRad, Hercules, CA). A similar approach for analyzing cytokines in BAL (bronchoalveolar lavage) fluid from asthmatics has been used previously[19 (link)]. Cytokines included in the analysis are listed in S1 Table.

Cytokine Profiling of CD8+ T-cells

Measurement of cytokines present in the supernatants of sorted CD8+CD146+ T-cells and CD8+CD146− T-cells stimulated with plate bound CD3/CD28 for 5 days. Comprehensive Clinical Biomarker Profiling

Hemoglobin and hematocrit were measured in EDTA-anticoagulated whole blood using a Sysmex XE-5000. Glucose, cholesterol, triglyceride, HDL, homocysteine, albumin, creatinine, fibrinogen, ferritin, and hsCRP were measured in plasma (Roche Cobas 6000 analyzer, Roche Diagnostics, Indianapolis, IN, USA). HbA1c values were determined by HPLC using a National Glycohemoglobin Standardization Program (NGSP)-certified instrument (D10) made by BioRad Laboratories (Hercules, CA, USA).
TNF-α and MCP-1 were measured in plasma using a Luminex bead array assay with the Multiplex kit (HCYTOMAG-60K-10, Millipore, Chicago, IL, USA) on a Bio plex 200 instrument using the Bio-plex 6.1 software (Bio-Rad, Hercules, CA, USA). Data were available for n = 168 participants. IL-6 was measured in plasma with a digital enzyme-linked immunosorbent assay (ELISA) using SiMOA technology (cat no. 101622, Quantrix, Billerica, MA, USA). IL-6 levels were available in n = 182.
