We used the Flow Cellect Mito Damage Kit and Annexin V-PE Apoptosis Detection Kit obtained from EMD Millipore in the United States. To perform the flow cytometric analysis and understand how cells responded to HAMLET, we seeded 100,000 to 130,000 cells per well. After one day, we treated these cells with different concentrations of HAMLET for 6 h. We then detached the cells, stained them with Annexin V-PE and 7-AAD, and analyzed them using the Guava Personal Cell Analysis Flow Cytometer and CytoSoft software (version 2.1.4; Guava; EMD Millipore, Burlington, MA, USA).

Apoptosis Analysis of A549 Cells

A total of 1 × 105 A549 cells was seeded in a 24-well plate and grown until 80% confluence. Four hundred microliters of CG-M, CG-Mblank, celastrol + G Rh2, celastrol (20 μM), or G Rh2 (8 μM) solutions were added to each well. After 8 h of incubation, the cells were rinsed thoroughly with ice-cold PBS and digested with 200 μL of trypsin to harvest the cell suspension, which was then transferred (50 μL) to a 96-well plate and incubated with an equal volume of Annexin V-PE Apoptosis detection kit (Guava, Merck-Millipore, MA) in the dark for 15 min. Cell apoptosis was measured by flow cytometry (Guava 6HT).

Annexin V-PE Apoptosis Assay in A549 Cells

Apoptosis of the A549 cells at prearranged time interval was determined using an Annexin V-PE apoptosis detection kit (Merck-Millipore) according to the manufacturer’s protocol. Briefly, the cells were trypsinized and then terminated rapidly by phosphate-buffered saline containing 10% fetal bovine serum. Next, 50 μL of the above solution was mixed with 50 μL Annexin V-PE and incubated for 15 minutes. The cells were analyzed immediately by flow cytometry (Guava 6HT; Merck-Millipore).
