Profiling Phenolics and Flavonoids in Edible Oils

Fourteen edible oils (sunflower, safflower, canola, soybean, cotton, grape, flax, avocado, chia, Inca inchi, perillartine, sesame, and rice bran oil) were purchased from different supermarkets in Hiroshima Prefecture, Japan, in 2016. Details are described in Table 1. Standard chemicals for the analysis of individual phenolic acids (gallic acid, protocatechuic acid, catechol, chlorogenic acid, p-hydroxybenzoic acid, vanillic acid, caffeic acid, syringic acid, vanillin, ferulic acid, sinapic acid, p-coumaric acid, benzoic acid, ellagic acid, and cinnamic acid), and flavonoids (esculetin, isoquercetin, myricetin, fisetin, morin, quercetin, luteolin, kaempferol, isohamnetin, apigenin, rhamnetin, and galangin), extracting solvents, and buffers were of analytical grade and purchased from Wako company, Tokyo, Japan.

Quantification of Polyphenols and Flavonoids

The total phenolic content was measured by a colorimetric assay using Folin-Ciocalteu's phenol reagent [20 ]. A total of 0.96 mL of distilled water and 0.1 mL of 50% Folin-Ciocalteu's phenol were added to 0.04 mL of the sample, which was diluted with distilled water and incubated for 3 min. Then, 0.2 mL of 10% Na2CO3 was added, the mixture was incubated for 1 h, and the absorbance was subsequently measured at 700 nm. The total polyphenol content was calculated from a standard curve generated using gallic acid (Sigma, St. Louis, MO, USA).
The total flavonoid contents were determined according to the method described by Moreno et al. [21 (link)]. A total of 10% aluminum nitrate 0.02 mL, 1 M potassium acetate 0.02 mL, and 80% ethanol 0.86 mL were mixed with 0.4 mL of each sample, and each mixture was then incubated for 40 min at room temperature. Absorbance was measured at 415 nm using a spectrophotometer (Optizen 2120 UV, Mecasys, Korea). The total flavonoid content was determined from a calibration curve using quercetin (Wako, Osaka, Japan) as a standard.

Phytochemical and Antioxidant Assays

All the chemicals used in this study were of laboratory grade. The Folin–Ciocalteu's phenol reagent (FCR), α-amylase and acarbose were obtained from Sigma Aldrich, St. Louis, MO, USA. Sulphuric acid, hydrochloric acid, aluminium chloride, sodium chloride, sodium carbonate, dimethyl sulphoxide, potassium acetate, disodium hydrogen phosphate dihydrate, and sodium dihydrogen phosphate dihydrate (Thermo-Fisher Scientific India) were purchased from Kathmandu, Nepal. quercetin and gallic acid (Hi-media Laboratories) were procured from the local city of Kathmandu, Nepal. The 1,1-diphenyl-2-picrylhydrazyl (DPPH), gallic acid and quercetin (Wako Pure Chemicals, Osaka, Japan), were purchased from the local city of Kathmandu, Nepal.

Prostanoid and Nucleotide Quantification

All prostanoids (PGE3, PGF, TXB3, PGE2, PGF, and TXB2), deuterated prostanoids (PGE2-d4, PGF-d4, and TXB2-d4), and EPA were purchased from Cayman Chemical Co. (Ann Arbor, MI). Calcium ionophore A23187 and indomethacin were purchased from Sigma Aldrich (St. Louis, MO). Adenosine 5′-triphosphate (ATP) disodium salt hydrate, 5′-adenylic acid (AMP), and sodium creatine phosphate hydrate were purchased from Tokyo Chemical Industry Co., Ltd. (Tokyo, Japan). Creatine kinase was purchased from Roche Applied Science (Tokyo, Japan). Dipyridamole, probenecid, quercetin, methotrexate, and folic acid were purchased from Wako Pure Chemical Industries (Osaka, Japan). MK571, celecoxib, candesartan, estradiol 17β-glucronide (E217βG), adenosine 3′, 5′-cyclic monophosphate (cAMP), and guanosine 3′, 5′-cyclic monophosphate (cGMP) were purchased from Sigma Aldrich (St. Louis, MO). All other chemicals were of the highest purity available.

Cell Proliferation Assays of Food Constituents

We examined the inhibitory activities of food constituents in cell proliferation assays. Test compounds used in the assays were as follows: curcumin (Wako or Sigma-Aldrich), zerumbone (Wako), genistein (Sigma-Aldrich), chalcone (Wako), corosolic acid (Wako), carnosol (Wako), ursolic acid (Wako), epigallocatechin (Sigma-Aldrich), isoflavone (Wako), apigenin (Wako), chrysin (Wako), flavanone (Wako), resveratrol (Wako), flavone (Wako), quercetin (Wako), daidzein (Wako), and clodronate (Sigma-Aldrich). These compounds were firstly dissolved with dimethyl sulfoxide (DMSO; Wako) and then appropriately diluted with RPMI1640 + 10% FCS to apply the assays. The final concentration of DMSO was adjusted not to exceed 1% at most in cell cultures.

Nitrotetrazolium Blue chloride (NBT) was obtained from Sinopharm Chemical Reagent Co., Ltd. (Beijing, China). Kanamycin and carbenicillin were purchased from Shanghai Xinya Pharmaceutical Co., Ltd. (Shanghai, China). Fetal bovine serum (BP) of no phage and low endotoxin was from Sijiqing Biological Engineering Materials Co., Ltd. (Hangzhou, China). All other reagents and solvents were analytical grade and used without further purification.
